Effects of Stellera chamaejasme L.on related cytokines and intestinal flora in adjuvant arthritis rats
Objective:To observe the effects of Stellera chamaejasme L.(SCL)on related cytokines and intestinal flora in adjuvant arthritis(AA)rats.Methods:The AA model was established by subcutaneous injection of 0.1 mL Freund's complete adjuvant into the right hind paw of male SD rats on the 1st,7th and 14th days.From the 8 th day,the positive group was given 12.86 mg/kg tripterygium wilfordii polyglycoside tablets by gavage,and the high,medium and low dose groups of SCL were given 32.97,16.49 and 8.24 g/kg respectively by gavage for 21 days.The protective effect of SCL on AA was evaluated by statistics of rat paw swelling and histopathology.Based on 16S rRNA sequencing,the effects of SCL on intestinal flora and inflammatory factors in AA rats were investigated by ELISA.Results:SCL could significantly improve the joint pathological symptoms of AA rats,and the paw swelling degree of rats in SCL administration group was significantly reduced.Compared with the model group,the levels of serum tumor necrosis factor-α(TNF-α),interleukin(IL)-6,IL-1 β,estradiol(E2)and rheumatoid factor(RF)in the high-dose SCL group were significantly decreased(P<0.01,P<0.05),and the level of progesterone(PROG)was significantly increased(P<0.01).The levels of TNF-α,IL-6 and RF in serum of SCL medium dose group were significantly decreased(P<0.01,P<0.05),and the level of E2 was significantly increased(P<0.01).The level of TNF-α in serum of SCL low dose group was significantly decreased(P<0.05).The serum levels of TNF-α,IL-6 and PROG in the positive group were significantly decreased(P<0.01).In terms of intestinal flora diversity,SCL significantly increased the relative abundance of bacteroidetes,bacteroides,S247 family,prevotaceae,bacteroidales,S247class,prevotclass,enterococcus class,lactobacillus class(P<0.05),and decreased the relative abundance of coccidiaceae,alostridium,lactobacillaceae,and frium(P<0.05).Conclusion:SCL may play an anti-inflammatory role by regulating the imbalance of intestinal flora,and then treat AA,and provides a theoretical basis for the development of new drugs for AA in clinical practice.
