腹针调控水、盐、糖代谢治疗慢性心理应激肺癌荷瘤小鼠实验

目的:证实肺癌的进展和慢性心理应激相关,腹针可调节糖代谢和水盐代谢平抚慢性心理应激创伤并抑制肺癌.方法:将7~8月龄雄性C57BL/6J小鼠按体质量随机分为4组,每组16只.动物分组如下:对照组:小鼠荷瘤,自由进食进水,不加任何干预;慢性心理应激组:小鼠荷瘤,通过隔天抓握小鼠后背皮肤10 s达到慢性心理应激效果,观察指标主要为毛发色泽的变浅;慢性心理应激+腹针组(以下简称腹针组):小鼠荷瘤,抓住小鼠后背皮肤并取穴腹部任脉气海、关元、中脘穴,脾经大横(双侧)、腹结(双侧)、府舍(双侧),每次针1个穴位,每个穴位针刺提插捻转10 s,平补平泻,隔天1次;慢性心理应激+环磷酰胺组(以下简称环磷酰胺组):小鼠荷瘤,抓住小鼠后背皮肤并腹腔注射环磷酰胺,50 mg/kg给药,隔5天1次.适应性喂养1周,施针或给环磷酰胺1周后,全部小鼠接种0.2 mL(5×106/mL)Lewis肺癌细胞于右侧前肢腋窝下,继续施针或给药1周瘤体萌出,再继续施针或给药1周取瘤称重,并检测糖代谢相关蛋白AKT2、GSK3β,水盐代谢相关蛋白AQP1、ATP1A1,抗凋亡蛋白MCL1的mRNA和蛋白表达,使用SPSS 22.0统计软件计算对比各组检测数据.结果:慢性心理应激小鼠局部毛发色泽变浅,腹针组肿瘤明显比慢性心理应激组及对照组的肿瘤生长缓慢(P<0.01),腹针组肿瘤糖代谢相关蛋白AKT2、GSK3β,水盐代谢相关蛋白AQP1、ATP1A1,抗凋亡蛋白MCL1 mRNA及蛋白表达较慢性心理应激组均显著降低(P<0.01).结论:肺癌的发展和慢性心理应激相关,腹针可以通过调节糖代谢通路AKT2/GSK3 β和水盐代谢相关蛋白AQP1、ATP1A1平抚慢性心理应激的伤害,降低抗凋亡蛋白MCL1蛋白的表达以抑制肺癌.
Abdominal acupuncture regulates the metabolism of water,salt and sugar to treat chronic psychological stress lung cancer bearing mice experiment
Objective:To confirm that the progression of lung cancer is related to chronic psychological stress,and abdominal acupuncture can regulate glucose metabolism and water and salt metabolism to calm chronic psychological stress trauma and inhibit lung cancer.Methods:Male C57BL/6J mice aged 7-8 months were randomly divided into 4 groups according to body weight,with 16 mice in each group.The animals were divided into the following groups:control group,mice bearing tumor,feeding and drinking freely without any intervention;Chronic psychological stress group,mice bearing tumor,by grasping the back skin of mice every other day for 10 s to achieve chronic psychological stress effect,the main observation index is hair color lightening;Chronic psychological stress+abdominal acupuncture group(referred to as abdominal acupuncture group):mice bearing tumor,hold the back skin of mice and select the abdominal Renmai Qihai,Guanyuan and Zhongwan points,spleen meridian Dahorizontal(bilateral),Fujie(bilateral),Fushe(bilateral),needle one point each time,each acupuncture point lifting and twisting for 10 s,flat supplement and flat diarrhea,once every other day;Chronic psychological stress plus cyclophosphoxide group(cyclophosphoxide group for short),mice bearing tumor,hold the back skin of mice and intraperitoneal injection of cyclophosphoxide,50 mg/kg dose,once every 5 days.After 1 week of adaptive feeding and 1 week of injection or cyclophospholide,all mice were inoculated with 0.2 mL(5×106/mL)Lewis lung cancer cells under the armpit of the right forelimb,and the tumors erupted after continued injection or administration for 1 week,and the tumors were taken and weighed after continued injection or administration for 1 week.The mRNA and protein expressions of glucose metabolism-related proteins AKT2 and GSK3 β,water and salt metabolism-related proteins AQP1 and ATP1A1,and anti-apoptosis protein MCL1 were detected.SPSS 22.0 statistical software was used to calculate and compare the detection data of each group.Results:The local hair color of mice under chronic psychological stress became lighter,and the tumor growth in the abdominal acupuncture group was significantly slower than that in the chronic psychological stress group and the control group(P<0.01).The tumor proteins related to glucose metabolism AKT2 and GSK3 β,and the proteins related to water and salt metabolism AQP1 and ATP1A1 in the abdominal acupuncture group were significantly slower than that in the chronic psychological stress group and the control group.The mRNA and protein expression of anti-apoptosis protein MCL1 were significantly decreased compared with chronic psychological stress group(P<0.01).Conclusion:The development of lung cancer is related to chronic psychological stress.Abdominal acupuncture can relieve the damage caused by chronic psychological stress by regulating glucose metabolism pathway AKT2/GSK3 β,water and salt metabolity-related proteins AQP1 and ATP1A1,reducing the expression of anti-apoptosis protein MCL1 and inhibiting lung cancer.
