基于TLR4/NF-κB信号通路探讨柴胡陷胸汤含药血清对脂多糖诱导人胃黏膜上皮GES-1细胞的作用机制

目的:探讨柴胡陷胸汤含药血清对脂多糖(LPS)诱导人胃黏膜上皮GES-1细胞Toll样受体4(TLR4)/核因子κB(NF-κB)信号通路的影响,以揭示该方治疗慢性胃炎可能的作用机制.方法:GES-1细胞分为4组:空白组:GES-1细胞+正常大鼠血清;模型组:GES-1细胞+LPS诱导+正常大鼠血清;柴胡陷胸汤组:GES-1细胞+LPS诱导+柴胡陷胸汤含药血清;吡咯烷二硫代甲酸铵(PDTC)组:GES-1细胞+LPS诱导+PDTC.运用MTT检测LPS的细胞毒性;运用CCK8法检测细胞活性;运用ELISA法检测细胞炎症因子白细胞介素(IL)-1β、IL-6水平;运用Western Blot法检测TLR4、NF-κB、p-NF-κB、核苷酸寡聚结合域样受体蛋白3(NLRP3)蛋白表达水平.结果:与空白组比较,模型组细胞炎症因子IL-1β、IL-6水平显著升高(P<0.01),与模型组比较,给予柴胡陷胸汤含药血清作用后,GES-1细胞IL-1β、IL-6水平显著降低(P<0.01);与空白组比较,模型组GES-1细胞TLR4、NLRP3蛋白表达及NF-κB磷酸化水平显著升高(P<0.01),与模型组比较,柴胡陷胸汤组GES-1细胞TLR4、NLRP3蛋白表达及NF-κB磷酸化水平显著降低(P<0.01).结论:柴胡陷胸汤可通过TLR4/NF-κB信号通路抑制LPS诱导的IL-1β和IL-6分泌,减轻GES-1细胞损伤.
Mechanism of Chaihu Xianxiong Decoction-containing serum on human gastric epithelial GES-1 cells induced by lipopolysaccharide based on TLR4/NF-κB signaling pathway
Objective:To investigate the effect of serum containing Chaihu Xianxiong Decoction(CHXXD)on TLR4/NF-κB signaling pathway induced by lipopolysaccharide(LPS)in human gastric epithelial GES-1 cells,and to reveal the possible mechanism of CHXXD in treating chronic gastritis.Methods:GES-1 cells were divided into the following four groups:blank group:GES-1 cells+normal rat serum;model group:GES-1 cells+LPS induction+normal rat serum;CHXXD group:GES-1 cells+LPS induction+serum with CHXXD;PDTC group:GES-1 cells+LPS induction+PDTC.Cell morphology was observed and cytotoxicity of LPS was detected by MTT;the effect of drug serum on GES-1 cell viability was detected by CCK8;the levels of IL-1β and IL-6 in each group were detected by ELISA;the protein expressions of TLR4,NF-κB,phosphorylated NF-κB and NLRP3 in all groups were detected by Western Blot.Results:Compared with the blank group,the levels of inflammatory cytokines IL-1β and IL-6 in model group were significantly increased(P<0.01).Compared with the model group,the levels of IL-1β and IL-6 in GES-1 cells were significantly decreased after the combined action CHXXD drug serum(P<0.01).Compared with the blank group,the protein levels of TLR4,phosphorylated NF-κB and NLRP3 in the model group were significantly increased(P<0.01).Compared with the model group,the levels of TLR4,phosphorylated NF-κB and NLRP3 protein in GES-1 cells were significantly decreased after combined treatment with CHXXD(P<0.0l).Conclusion:CHXXD can inhibit the secretion of IL-1β and IL-6 induced by LPS through TLR4/NF-κB signaling pathway,and alleviate the injury of GES-1 cells.
