Research on the mechanism of hUC-MSCs-Exos combined with Compound Muji Granules inhibiting the proliferation of human liver cancer cell SMMC-7721 based on metabolomics
Objective:To investigate the mechanism of human umbilical cord mesenchymal stem cells(hUC-MSCs)derived exosomes(hUC-MSCs-Exos)combined with Compound Muji Granules on the proliferation and apoptosis of human liver cancer cell line SMMC-7721 based on metabolomics.Methods:Exosomes were extracted from hUC-MSCs.The proliferation inhibition rate and apoptosis necrosis of human liver cancer cell line SMMC-7721 were detected by detecting the Compound Muji Granules,extracellular vesicles,and combined administration,to verify the efficacy differences before and after combined administration.Key targets in pathways such as glycolysis,inflammation,and oxidative stress were detected by ELISA.The mechanism of action was studied by using cell metabolomics combined with UPLC-Q-TOF-MS technology.Results:Compound Muji Granules can significantly inhibit the proliferation of human liver cancer cell line SMMC-7721,and the inhibitory effect of exosomes is not significant,but the combined application of the two has an enhanced effect.The combination of Compound Muji Granules and exosomes can recall targets on multiple pathways before and after treatment.Conclusion:The mechanism of enhanced efficacy of Compound Muji Granules combined with hUC-MSCs-Exos may be related to lysine degradation,fatty acid degradation,fatty acid elongation,nitrogen metabolism,caffeine metabolism.
