Study on the inhibition of isoproterenol-induced myocardial hypertrophy in mice by acupuncture at Neiguan acupoint(PC 6)
Objective:To observe the effect of acupuncture on the pathological structure of myocardial tissue in myocardial hypertrophic mice.Methods:Sixty male C57BU6 mice were randomly divided into control group,model group,acupuncture group,sham group and medicine group,with 12 mice in each group.Mice were given subcutaneous injection of isoproterenol(ISO)(25 mg·kg-1·d-1)to prepare a model of myocardial hypertrophy for 14 consecutive days,except for the control group.After the modeling,the mice in the acupuncture group were treated with acupuncture bilateral Neiguan acupoint(PC 6)for 30 min/time.The sham group selected a certain position on the tail of the mouse(not corresponding to any traditional acupuncture point)for acupuncture,30 min/time.The medicine group was given 10 mg/kg propranolol by gavage once a day.All groups were treated for 14 days.After the end of treatment,the heart rate of mice in each group was measured,and LVAWd,LVPWd,LVAWs,LVPWs,and ejection fraction(EF,%)were detected by echocardiography.Calculate the heart-to-weight ratio(cardiac weight/tibial length ratio).HE staining and Masson staining were used to observe the pathological structural changes of myocardium.Results:Compared with the control group,the values of heart rate,LVAWs,LVPWs,LVPWd,LVAWd and cardiac weight ratio in the model group and the sham group were increased(P<0.01,P<0.05).Compared with the model group,the values of heart rate,LVAWs,LVPWd,LVAWd and heart-to-weight ratio were decreased in the acupuncture group and the medicine group(P<0.01,P<0.05).The pathological morphology of myocardium was observed by HE staining and Masson staining,and the degree of myocardial damage in the acupuncture group and the mdeicine group was milder than that in the model group.Conclusion:Acupuncture on Neiguan acupoint(PC 6)can slow down heart rate,inhibit myocardial fibrosis,improve cardiac hypertrophy and reduce myocardial damage.
