芒柄花黄素的急性和亚急性毒性评价

摘要
目的:研究芒柄花黄素经口给药对小鼠的急性和亚急性毒性.方法:ICR小鼠,雌雄各半,单次灌胃超大剂量芒柄花黄素(2 000和4 000 mg/kg)后观察随后14 d的动物状态.在亚急性毒性试验中,各组小鼠连续灌胃芒柄花黄素(50、100、250、500、1 000 mg/kg)14d后,观察小鼠体质量、各脏器指数、血液学、肝功能、肾功能和组织病理学变化.结果:在急性毒性实验中,芒柄花黄素对小鼠无明显毒性和致死.在亚急性毒性实验中,芒柄花黄素对小鼠体质量、血液学参数无明显影响.芒柄花黄素500和1 000 mg/kg组雌性小鼠血浆肌酐水平较正常组显著升高(P<0.05,P<0.01),芒柄花黄素1 000 mg/kg组雌性小鼠卵巢指数较正常组显著增加(P<0.01),但组织病理学显示均无明显变化.结论:芒柄花黄素经口毒性实验无可见有害作用水平值>4 000 mg/kg,具有非常高的安全性,但提示长期高剂量芒柄花黄素对雌性小鼠卵巢和肾脏可能有潜在影响.
Abstract
Objective:To study the acute and sub-acute toxicity of formononetin orally administered in mice.Methods:Male and female ICR mice were given a single dose of formononetin(2 000 and 4 000 mg/kg)to observe the toxicity effects for 14 d.In the sub-acute toxicity test,formononetin(50,100,250,500,1 000 mg/kg)was orally administered continuously for 14 d,the body weight,organ index,hematology,liver and kidney function,and histopathology were observed.Results:In the acute toxicity test,formononetin had no obvious toxicity or death in mice.In the sub-acute toxicity test,formononetin did not affect body weight and hematological parameters of mice.Plasma creatinine level of female mice was significantly increased in 500 and 1 000 mg/kg of formononetin groups than the normal group(P<0.05,P<0.01),and the ovarian weight index of female mice was significantly increased in 1 000 mg/kg of formononetin group than the normal group(P<0.01),but there was no significant histopathological change.Conclusion:The no observed adverse effect level of formononetin is greater than 4 000 mg/kg,which is very safe,but the long-term and high dose of formononetin may have potential effects on ovary and kidney of female mice.
