山蜡梅叶颗粒通过调控Th1/Th2平衡缓解变应性鼻炎大鼠炎症反应的机制

目的:探讨山蜡梅叶颗粒缓解变应性鼻炎(AR)的作用及相关机制.方法:60只SD大鼠随机分成空白组、模型组、阳性药组(鼻炎灵片,0.432 g/kg)、山蜡梅叶颗粒低、中、高剂量组(山蜡梅叶颗粒,3.15、6.30、12.60 g/kg).除空白组外,其余各组采用卵清蛋白(OVA)滴鼻建立AR模型.对各组大鼠鼻部症状行为学进行评分;给药结束后取血,进行血细胞分析并通过吉姆萨染色对全血中嗜酸粒细胞计数分析;ELISA法检测血清中免疫球蛋白E(IgE)和组胺(HIS)含量及大鼠鼻腔灌洗液中肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-10、IL-4、γ-干扰素(IFN-γ)水平;HE染色观察大鼠鼻中隔鼻黏膜组织病理变化;Western Blot和RT-qPCR分别检测鼻黏膜中信号转导和转录激活因子(STAT)6、STAT4、GATA结合蛋白3(GATA3)、T细胞表达的T盒(T-bet)、p65、核因子κB抑制因子α(IκB-α)蛋白及mRNA表达情况.结果:与空白组比较,模型组大鼠喷嚏和挠鼻等行为学分数显著增加(P<0.01),血液中白细胞计数显著升高(P<0.01),血小板计数显著下降(P<0.01),嗜酸性粒细胞数升高,血清中IgE和HIS水平、鼻腔灌洗液中TNF-α、IL-10和IL-4含量均显著上升(P<0.01,P<0.05),鼻黏膜组织中STAT6、STAT4、GATA3、T-bet、p65和IκB-α 的表达差异显著(P<0.01):鼻黏膜组织间水肿明显.与模型组比较,山蜡梅叶颗粒各剂量组给药后上述行为学指标均改善,大鼠行为学评分、白细胞计数显著降低(P<0.01),嗜酸性粒细胞数量减少,血清中IgE和HIS水平显著降低(P<0.01),山蜡梅叶颗粒高剂量组鼻腔灌洗液中TNF-α、IL-10、IL-4含量显著降低(P<0.05,P<0.01),IFN-γ含量显著增加(P<0.01);鼻黏膜组织中STAT6、GATA3、T-bet和p65的表达都有着显著差异(P<0.05),各给药组大鼠鼻黏膜充血、水肿、炎细胞浸润减轻.结论:山蜡梅叶颗粒对AR大鼠的炎症具有明显的改善作用,其机制可能与调节辅助性T细胞(Th)1/Th2免疫平衡有关.
Mechanism of alleviating inflammation reaction in rats with allergic rhinitis by regulating the Th1/Th2 balance with Shanlameiye Granules
Objective:To investigate the role and mechanism of Shanlameiye Granules in alleviating allergic rhinitis(AR).Methods:Sixty SD rats were randomly divided into normal control group,model control group,positive drug group(Rhinitis Spirit Tablet,0.432 g/kg),low,medium,and high dose groups of Shanlameiye Granules(Shanlameiye Granules,3.15,6.30,12.60 g/kg).Except for the normal control group,the model groups used ovalbumin(OVA)systemic and local attack methods to establish a rat model of AR.The nasal symptomatic behavior of rats was scored.Blood was taken after administration,blood cell analysis was performed and eosinophil count analysis in whole blood was performed by Giemsa staining.The contents of immunoglobulin E(IgE)and histamine(HIS)in serum and TNF-α,IL-10,IL-4 and IFN-α in rat nasal lavage solution were detected by ELISA.HE staining observed the histopathological changes of nasal mucosa in rats.Western Blot and RT-qPCR method detected STAT6,STAT4,GATA3,T-be,p65,IκB-α protein and mRNA expression in the nasal mucosa.Results:Compared with the blank control group,the behavioral scores of sneezing and nose scratching in the model group were significantly increased(P<0.01),the number of leukocytes in the blood significantly increased(P<0.01),the number of platelet in the blood significantly decreased(P<0.01),and the number of eosinophils in the blood increased,and the levels of IgE and HIS in serum and TNF-α,IL-10 and IL-4 in nasal lavage fluid were significantly increased(P<0.01,P<0.05),the expression of STAT6,STAT4,GATA3,T-be,p65,and IκB-α in nasal mucosal tissues were significantly different(P<0.01);nasal mucosal intertissue edema was pronounced.Compared with the model group,the above indexes were significantly improved in each dose group of Shanlameiye Granules,and the behavioral scores,the number of leukocytes significantly decreased(P<0.01),and the number of eosinophils decreased,serum IgE and HIS levels significantly decreased(P<0.01),TNF-α,IL-10 and IL-4 contents in nasal lavage solution in high dose group of shanlameiye Granules were significantly reduced(P<0.05,P<0.01),the content IFN-γ significantly increased(P<0.01),the expression of STAT6,GATA3,T-bet and p65 in nasal mucosal tissues were significantly different(P<0.05),and nasal mucosal hyperemia,edema and inflammatory cell infiltration in rats in the administration group were reduced.Conclusion:Shanlameiye Granules have a significant effect on the inflammation of AR rats,and the mechanism may be related to the regulation of Th1/Th2 immune balance.
