中药及天然药物中黄酮类成分抗血小板聚集作用的研究进展

血小板的激活、黏附和聚集在阻塞性血管疾病的发病机制中具有重要的作用.黄酮类成分是普遍存在于植物中的多酚类化合物,能够通过不同生物学途径产生抗血小板聚集的作用.文章通过对血小板聚集机制及黄酮类成分体内吸收与代谢的分析,阐述了黄酮类成分及其体内中间代谢产物抗血小板聚集效应和其相关的作用机制,并对当前有关黄酮类成分抗血小板聚集临床研究现状进行了总结.由此认为,尽管目前黄酮类成分可以通过干预血小板活化的相关靶点达到抗血小板聚集作用,但还需要进一步明确其关键环节,从不同角度深入探讨中药及复方中黄酮类成分和其中间代谢产物抗血小板聚集的确切机制,将有助于抗血小板聚集新药研发,也为临床血栓性疾病的中医药治疗提供重要参考.
Platelet activation,adhesion and aggregation have an important role in the pathogenesis of obstructive vascular diseases.Flavonoids are polyphenolic compounds commonly found in plants and can have anti-platelet aggregation effects through differently biological pathways.In this paper,we summarize the anti-platelet aggregation effects of flavonoids and their intermediate metabolites in vivo through the analysis of platelet aggregation mechanisms and absorption and metabolism of flavonoids in vivo.The current status of clinical studies on the anti-platelet aggregation of flavonoids in plants is also demonstrated.It is concluded that further study of key mechanisms of anti-platelet aggregation of flavonoids is needed although the flavonoids can achieve anti-platelet aggregation effects by interfering with the main targets in the process of platelet activation.The in-depth investigation of the exact mechanism of anti-platelet aggregation of flavonoids and their intermediary metabolites in traditional Chinese medicine and formulas in different points will contribute to the development of new anti-platelet aggregation drugs,and also provide an important guideline for the clinical thrombotic diseases in traditional Chinese medicine.
