摘要

目的:观察瘀浊脂消丸对高脂饲料诱导的非酒精性脂肪性肝炎(NASH)大鼠的治疗作用,并基于肠道菌群变化探讨其作用机制.方法:将40只雄性SD大鼠随机均分为空白(Con)组、模型(MOD)组、瘀浊脂消丸(YZZXP)组和双歧杆菌(Prob)组.高脂饮食喂养12周建立大鼠NASH模型.给药12周后检测大鼠肝功能及血脂各项指标;取结肠内容物做16S rDNA检测;取肝组织进行HE染色和油红O染色,并且检测其甘油三酯(TG)含量.结果:与Con组比较,MOD组大鼠丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、总胆汁酸(TBA)、总胆固醇(TC)、甘油三酯(TG)和低密度脂蛋白胆固醇(LDL-C)显著升高(P<0.05),而高密度脂蛋白胆固醇(HDL-C)显著降低(P<0.05),肝脏病理形态恶化,TG和NAS评分显著升高(P<0.05),Oscillospira和Roseburia等8个菌属显著上调(P<0.05),Bifidobacterium和Lachnospira等6个菌属显著下调(P<0.05);与MOD组比较,YZZXP组ALT、AST、ALP、TBA、TC、TG和LDL-C显著降低(P<0.05),而HDL-C显著升高(P<0.05),肝脏病理形态好转,Akkermansia和Turicibacter等8个菌属显著上调(P<0.05),Ralstonia和Paraprevotella等1o个菌属显著下调(P<0.05).结论:瘀浊脂消丸可以抑制Roseburia等有害菌属繁衍,增加Akkermansia等有益菌属来调节代谢途径,调控血脂治疗高脂饮食诱导的NASH.

Abstract

Objective:To observe the therapeutic effect of Yuzhuo Zhixiao Pill on rats with nonalcoholic steatohepatitis(NASH)induced by high fat diet,and to explore its mechanism based on the changes of intestinal flora.Methods:A total of 40 male SD rats were randomly divided into blank(Con)group,model(MOD)group,Yuzhuo Zhixiao Pill(YZZXP)group and bifidobacterium(Prob)group.NASH model was established after 12 weeks of high fat diet.The indexes of liver function and blood lipid were detected after 12 weeks of administration.Colon contents were collected for 16S rDNA detection.Liver tissue was stained with HE and oil red O,and triglyceride(TG)content was detected.Results:Compared with Con group,ALT,AST,ALP,TBA,TC,TG and LDL-C in the MOD group were significantly increased(P<0.05),while HDL-C in the MOD group was decreased(P<0.05),liver pathological morphology was deteriorated,TG and NAS score were significantly increased(P<0.05),and 8 bacteria genera such as Oscillospira and Roseburia were significantly up-regulated in MOD group(P<0.05),Bifidobacterium and Lachnospira were significantly down-regulated in MOD group(P<0.05).Compared with MOD group,ALT,AST,ALP,TBA,TC,TG and LDL-C in the YZZXP group were significantly decreased(P<0.05),while HDL-C in the YZZXP group was increased(P<0.05),YZZXP group improved liver pathological morphology,and significantly up-regulated 8 bacteria genus such as Akkermansia and Turicibacter(P<0.05),Ralstonia and Paraprevotella were significantly down-regulated(P<0.05).Conclusion:Yuzhuo Zhixiao Pill can inhibit the proliferation of harmful bacteria such as Roseburia,increase the beneficial bacteria such as Akkermansia,regulate metabolic pathway,regulate blood lipid in the treatment of high-fat diet-induced NASH.
