摘要

目的:研究香连止泻片对溃疡性结肠炎(UC)小鼠辅助性T细胞17(Th17)/调节性T细胞(Treg)免疫轴的影响.方法:噁唑酮诱导复制UC小鼠模型,将小鼠随机分为空白组,模型组,美沙拉秦组,香连止泻片低、中、高剂量组,每组8只.每日进行疾病活动指数(DAI)评分;统计胸腺指数;HE染色观察结肠组织病理形态并进行结肠黏膜损伤系数(CMDI)评分;ELISA检测结肠组织白细胞介素(IL)-10、转化生长因子-β1(TGF-β1)含量;Western Blot检测结肠组织叉头框蛋白P3(FOXP3)、IL-17A蛋白;流式细胞术检测血浆中Th17细胞和Treg细胞.结果:与空白组比较,模型组小鼠DAI评分、CMDI评分、TGF-β1含量、IL-17A蛋白表达和Th17/Treg比值显著升高(P<0.01,P<0.05),胸腺指数、IL-10含量和FOXP3蛋白表达显著下降(P<0.01,P<0.05),大量炎症细胞浸润至黏膜下层,黏膜下层水肿增厚,隐窝扭曲并萎缩且数量减少.与模型组比较,各给药组小鼠第7天DAI评分显著下降(P<0.01,P<0.05),CMDI评分、TGF-β1含量和Th17/Treg比值显著下降(P<0.01,P<0.05),香连止泻片高剂量组IL-17A蛋白表达显著下降(P<0.05),香连止泻片低剂量组胸腺指数显著升高(P<0.01),香连止泻片高剂量组IL-10含量显著升高(P<0.05),香连止泻片中剂量组FOXP3蛋白表达显著升高(P<0.05),各给药组小鼠结肠黏膜损伤有较好的缓解.结论:香连止泻片对UC小鼠有一定的治疗作用,其机制可能与影响Th17/Treg免疫轴相关.

Abstract

Objective:To investigate the effect of Xianglian Zhixie Tablet on Th17/Treg immune axis in ulcerative colitis(UC)mice.Methods:OXA-induced UC mice were randomly divided into blank group,model group,Mesalazine group,and Xianglian Zhixie Tablet low,medium,and high dose groups,8 mice in each group.The DAI score of mice was evaluated daily.The changes of thymus index of mice were analyzed.HE staining was used to observe the changes of colonic tissue pathology in mice,and to score the CMDI.ELISA was used to detect the changes of IL-10 and TGF-β1 content in colonic tissue of mice.Western Blot was used to detect the expression of FOXP3 and IL-17A protein in colonic tissue of mice.Flow cytometry was used to detect the proportion of Th 17 cells and Treg cells in mouse plasma.Results:Compared with the blank group,the DAI score,CMDI score,TGF-β1 content,IL-17A protein expression and Th17/Treg ratio in the model group were significantly increased(P<0.01,P<0.05),while the thymus index,IL-10 content and FOXP3 protein expression were significantly decreased(P<0.01,P<0.05).A large number of inflammatory cells infiltrated into the submucosa,the submucosa was edematous and thickened,and the cryps were distorted and atrophied and decreased in number.Compared with the model group,the DAI score on the 7th day,CMDI score,TGF-β1 content and Th17/Treg ratio of the mice in each administration group were significantly decreased(P<0.01,P<0.05).The expression of IL-17A protein in the high-dose group of Xianglian Zhixie Tablet was significantly decreased(P<0.05),and the thymus index in the low-dose group of Xianglian Zhixie Tablet was significantly increased(P<0.01),and the content of IL-10 in the high-dose group of Xianglian Zhixie Tablet was significantly increased(P<0.05),and the expression of FOXP3 protein in the high-dose group of Xianglian Zhixie Tablet was significantly increased(P<0.05).The colonic mucosal injury of mice in each treatment group was alleviated.Conclusion:Xianglian Zhixie Tablet have a certain therapeutic effect on UC mice,and its mechanism may be related to Th17/Treg immune axis.
