健脾润肤汤对人角质形成细胞丝聚蛋白和Caspase-14表达的影响

摘要
目的:观察健脾润肤汤对人角质形成细胞炎症状态下丝聚蛋白(FLG)、半胱氨酸天冬氨酸蛋白酶-14(Caspase-14)及相关细胞因子表达的影响.方法:应用Western Blot和ELISA检测白细胞介素(IL)-4刺激前、后人角质形成细胞FLG、Caspase-14、IL-1b、IL-13、IL-17、肿瘤坏死因子-α(TNF-α)的表达差异.结果:经健脾润肤汤含药血清高浓度组孵育后FLG及Caspase-14蛋白表达均较空白组显著升高(P＜0.05).与模型组比较,高浓度组IL-1b、TNF-α表达显著升高(P＜0.05),IL-13、IL-17表达显著降低(P＜0.05);低浓度组IL-13、IL-17表达显著降低(P＜0.05),TNF-α表达显著升高(P＜0.05).高浓度组IL-17表达较低浓度组显著升高(P＜0.05).结论:健脾润肤汤含药血清可以提高炎症环境下人角质形成细胞FLG和Caspase-14的表达,增强皮肤屏障功能;进而减少特应性皮炎发病中细胞因子的进一步产生,提高IL-1b、TNF-α表达,降低IL-13、IL-17水平,修复Th1/Th2平衡,减轻炎症程度.
Abstract
Objective:To observe the effects of Jianpi Runfu Decoction on the expressions of filaggrin(FLG),Caspase-14 and related cytokines in keratinocytes with inflammation.Methods:The expression differences of FLG,Caspase-14,interleukin(IL)-1b,IL-13,IL-17 and TNF-α in keratinocytes before and after stimulation of IL-4,were detected by Western Blot and ELISA.Results:The expressions of FLG and Caspase-14 in keratinocytes were higher than those in the blank group after incubation with the low concentration medicated serum of Jianpi Runfu Decoction(P＜0.05).Compared with the model group,the expression of IL-1b and TNF-α in the high concentration group was significantly increased(P＜0.05),while the expression of IL-13 and IL-17 was significantly decreased(P＜0.05),the expression of IL-13 and IL-17 in the low concentration group was significantly decreased(P＜0.05),while the expression of TNF-α was significantly increased(P＜0.05).The expression of IL-17 in the high concentration group was significantly higher than that in the low concentration group(P＜0.05).Conclusion:The serum containing Jianpi Runfu Decoction can increase the expression of FLG and Caspase-14 in keratinocytes with inflammation,enhance the skin barrier function and reduce the cytokines relate to AD.It could increase the expression of IL-1b and TNF-α,while reduce IL-13 and IL-17 levels,the balance between Th1/Th2 would be repaired,and the degree of inflammation can be reduced.
