Effect of different administration modes of vitamin B6 on the pharmacokinetics of donepezil hydrochloride in rats
OBJECTIVE To study the effect of tail vein injection and gavage administration of vitamin B6 on the pharmacoki-netics of donepezil hydrochloride in rats.METHODS Totally 18 male SD rats were randomly and equally divided into donepezil hydrochloride group,tail vein vitamin B6 co-administration group and gavage vitamin B6 co-administration group.The blood con-centration of drug was determined by ultra-high performance liquid chromatography-fluorescence method,and the drug concentration-time curve was drawn.The pharmacokinetic parameters were fitted with DAS 2.0 software and statistically ana-lyzed using SPSS 26.0 software.RESULTS Compared with donepezil hydrochloride group,the tail vein vitamin B6 co-administration group had significantly longer MRT0-t,MRT0-∞ and t1/2z(P<0.01),significantly greater Vz/F(P<0.01),and sig-nificantly lower Cmax(P<0.01);the gavage vitamin B6 co-administration group had significantly lower AUC0-t,AUC0-∞,and Cmax(P<0.01),MRT0-t,MRT0-∞ and t1/2z were significantly prolonged(P<0.01,P<0.05),and CLz/F and Vz/F were significantly increased(P<0.01).CONCLUSION Both tail vein injection and intragastric administration of vitamin B6 can reduce the plasma concentration of donepezil hydrochloride,slow down the metabolism of donepezil hydrochloride,and prolong its action time in rats.
