Spectrum-effect relationship in regulate Qi and relieve pain effect of medicine flavor compatibility and dif-ferent polar parts of Xiangguo Jianxiao Priscription
OBJECTIVE To study the relationship between the fingerprint of Xiangguo Jianxiao Prescription and its regulat-ing qi and relieving pain effect,to explore the rationality of its compatibility,and to clarify the material basis of regulating qi and relieving pain of Xiangguo Jianxiao Prescription combined with the spectrum-effect relationship of different polar parts.METHODS Ultra performance liquid chromatography(UPLC)was used to establish the fingerprints of flavor compatibility and different polar parts of Xiangguo Jianxiao prescription.Small intestine charcoal advancing method and acetic acid twisting test were used to investigate the effects of Xiangguo Jianxiao Prescription flavor compatibility and regulating Qi and relieving pain of different polar parts.Grey correlation degree and Pearson bivariate correlation analysis were used to analyze the spectrum-effect relation-ship.RESULTS Ten common peaks were identified by the fingerprint of Xiangguo Jianxiao Prescription flavor compatibility.The results showed that the alcohol extract group of Xiangguo Jianxiao Prescription had the best effect of regulating Qi and reliev-ing pain,followed by Valeriana jatamansi and Amomum tsao-ko group,Valeriana jatamansi and Aucklandia lappa group,and the effect of Amomum tsao-ko and Aucklandia lappa group was poor.By combining the two analysis methods of gray correlation and Pearson bivariate correlation,it was found that the compounds represented by peaks 2,4,5,6,9 and 10 were strongly corre-lated with the regulating Qi and relieving pain effect of Xiangguo Jianxiao Prescription,and three components were identified as chlorogenic acid(peak2),3,5-O-dicafeylquinic acid(peak 4),4,5-O-dicaffeoylquinic acid(peak5),and their chromatographic peaks were identified.Spectrum-effect correlation results of different polar parts of Xiangguo Jianxiao Prescription for regulating Qi and relieving pain showed that the compounds represented by peaks X31,X32,X33,X34 and X28 were highly correlated.The retention time and comparison of standards showed that Xiangguo Jianxiao prescription had a peak(5)and a peak(X31)in differ ent polar parts,which was 4,5-O-dicaffeoylquinic acid.Peak 6 and peak X32 were the same peak,not identified.CONCLUSION The regulating Qi and relieving pain effect of Xiangguo Jianxiao prescription is the result of the joint action of Valeriana jatamansi Amomum tsao-ko and Aucklandia lappa.4,5-O-dicaffeoylquinic acid and peak 6(X32)are the main mate-rial basis of Xiangguo Jianxiao prescription for regulating Qi and relieving pain.
