Effects of plantago-thistle compatibility on blood glucose and intestinal flora in diabetic mice
OBJECTIVE To explore the effects of plantago-thistle prescription on blood sugar and intestinal flora in diabetic mice.METHODS A diabetic mouse model was established by injecting alloxan into the tail vein,and after successful modeling,plantago-giant thistle prescription(18,12,8 g·kg-1)was given as intervention.Daily water intake,food intake,fasting blood glucose(FBG)and body mass were recorded after continuous administration for 3 weeks.The contents of glucose tolerance(OGTT),fasting serum insulin(INS)and hemoglobin a1c(HbA1c)of the mice in each group were measured after 3 weeks of administration.The pathological changes of liver and pancreas tissues of the mice in each group were observed by HE staining.16S rRNA high-throughput gene sequencing was used to analyze the changes of intestinal flora in each group of mice after drug treatment.RESULTS Compared with the model group,there were no significant changes in body mass,water intake and food intake in plantago and thistle combined prescription group(P<0.05),but FBG level decreased(P<0.05),OGTT and INS con-tents increased(P<0.05),the cytoplasmic vacuolation of liver cells was significantly reduced,the pathological damage of pancre-atic cells was alleviated,and the cell structure was more complete.In Alpha diversity,Shannon index and Simpson index were increased in psyllium-thistle high-dose group(P<0.01).In Beta diversity,the compatibility of psyllium-thistle group could change the growth and distribution of some intestinal flora in diabetic mice.At the phylum level,the community composition and structure of Bacteroidetes,Firmicutes and Proteobacteria were regulated.At the genus level,the composition and proportion of Bacteroides and Prevotella were adjusted.CONCLUSION The combination of plantago and thistle can regulate the structure and relative abundance of intestinal flora in diabetic mice,reduce the damage of hepatocytes and pancreatic cells,and improve the insu-lin resistance,which plays a hypoglycemic role.
