Effect of hordenine on melanin synthesis in B16 cells and mechanism research
OBJECTIVE To study the effect of hordenine on melanin synthesis process in B16 cells and its mechanism of action.METHODS Western blot was used to detect the expression of TYR,TRP1 and TRP2 in B16 cells after the treatment with hordenine(50,100,200 μmol·L-1)to determine the impact on the melanin synthesis process.After PKA agonist treatment,the correlation between the effect of hordenine on melanin synthesis process in B16 cells and cAMP/PKA/CREB/MITF signaling pathways.RESULTS Hordenine(50,100,200 μmol·L-1)could inhibit the protein expression of c AMP,PKA,p-CREB and MITF in melanoma B16 cells group,thus inhibiting TYR,TRP1 and TRP2 expression during melanin synthesis process.CONCLUSION Hordenine can inhibit melanin synthesis process in B16 cells,and the mechanism may be related to inhibiting cAMP/PKA/CREB/MITF signaling pathways.
