基于UPLC-LTQ-Orbitrap-MS技术分析越鞠丸水提物的化学成分及入血成分

目的:系统分析越鞠丸水提物的化学成分及入血成分,为阐明越鞠丸抗糖尿病的药效物质基础提供依据。方法:基于UPLC-LTQ-Orbitrap-MS技术,结合对照品、碎片离子信息、数据库及相关文献信息,运用Xcalibur 2。1软件鉴定化学成分;通过比对空白血清、越鞠丸含药血清,分析入血成分;结合PharmMapper、SwissTargetPrediction数据库筛选越鞠丸入血成分治疗糖尿病的作用靶点,Cytoscape软件做网络拓扑分析并筛选关键靶点,使用Autodock Vina软件对代表性入血成分与关键靶点进行分子对接。结果:越鞠丸水提物中共鉴定出116种化合物,包括黄酮类、环烯醚萜类、有机酸类、核苷类等,并对化合物的药材来源进行归属,在此基础上,发现34种成分可入血;越鞠丸治疗糖尿病的关键靶点37个,咖啡酸、栀子苷、黄芩苷、小檗碱与PPARγ、AKT1、TNF的对接结果良好(结合能<-20kJ·mol-1)。结论:通过系统分析越鞠丸水提物的化学成分及入血成分,发现34种入血成分可能是越鞠丸抗糖尿病潜在的有效成分。
Identification of chemical constituents and blood components of aqueous extracts of Yueju Pills by UPLC-LTQ-Orbitrap-MS
OBJECTIVE To systematically analyze the chemical constituents and blood components of aqueous extract of Yueju Pills(YP)and provide rationales for elucidating its antidiabetic pharmacological mechanism.METHODS Based upon UPLC-LTQ-Orbitrap-MS technology,in conjunctions with reference substances,fragment ion information,databases and related literature information,chemical components of YP were identified by Xcalibur 2.1 software.And its blood components were analyzed by comparing blank and drug-containing sera.The databases of PharmMapper and SwissTargetPrediction were uti-lized for screening the targets of blood components of YP for treating diabetes mellitus(DM),Cytoscape software was employed for performing network topology analysis and screening key targets.And Autodock Vina software was utilized for performing molecular docking between representative blood components and key targets.RESULTS A total of 116 constituents were identi-fied in aqueous extract of YP,including flavonoids,iridoids,organic acids and nucleosides,etc.The sources of Chinese medi-cines for all compounds in aqueous extract of YP were assigned.As a result,34 blood components were detected.There were 37 key therapeutic targets of YP and the docking results of caffeic acid,gardenoside,baicalin and berberine with PPARγ,AKT1 and TNF were excellent(binding energy<-20 kJ·mol-1).CONCLUSION By systematically analyzing the chemical constituents and blood components of aqueous extract of YP,34 blood components may be the potential active antidiabetic components of YP.
