目的:基于斑马鱼炎症模型筛选荔枝草抗炎药效物质。方法:运用UPLC-Q-TOF-MS/MS技术表征荔枝草95%、70%、50%、30%乙醇和水提取部位的化学成分质谱信息,确定共有成分;建立斑马鱼炎症模型,以TNF-α、IL-6以及IL-10表达量为指标评价各部位抗炎活性;利用皮尔森相关系数和灰色关联度分析共有成分峰面积与抗炎活性指标的相关性,筛选最佳抗炎活性部位及相关药效物质,并进行实验验证。结果:95%乙醇部位抗炎活性最好。5个提取部位有22个共有成分,其中高车前苷、高车前素、异鼠李素、木犀草素、芹菜素、鼠尾草酚和6-羟基木犀草素为荔枝草抗炎潜在药效物质。对前6个成分进行验证,结果显示均能显著降低斑马鱼炎症模型TNF-α、IL-6的表达,促进IL-10的表达(P<0。01)。结论:荔枝草抗炎药效物质的阐明为其质量评价、资源综合开发利用奠定了基础。
Screening for material basis of anti-inflammatory effect of Salvia plebeia based upon zebrafish inflamma-tion model
OBJECTIVE To screen for the anti-inflammatory substance of Salvia plebeia based upon zebrafish inflammation model.METHODS The chemical constituents of 95%,70%,50%,30%ethanol extraction parts and water extraction parts of Salvia plebeia were characterized by UPLC-Q-TOF-MS/MS for determining the common component Salvia plebeia.And the inflammatory model of zebrafish was established for evaluating the anti-inflammatory activity of each extract fraction according to the expression levels of tumor necrosis factor-alpha(TNF-α),interleukin-6(IL-6)and interleukin-10(IL-10).The correlation between peak area of common components in characterization map and anti-inflammatory activity index was established by Pear-son's bivariate analysis and grey correlation degree and anti-inflammatory quality markers(Q-markers)of Salvia plebeia was veri-fied.RESULTS 95%ethanol extract had the best anti-inflammatory activity.A total of 22 common components were identified from 5 extract sites.Homoplantaginin,hispidulin,isorhamnetin,luteolin,apigenin,camosol and 6-hydroxyluteolin were identi-fied as potential anti-inflammatory substances of Salvia plebeia,among which the first 6 easily obtained components were selected for anti-inflammatory activity verification.And the results showed that they could significantly lower the expressions of TNF-αand IL-6 and promote the expression of IL-10 in Zebrafish inflammatory model(P<0.01).CONCLUSION The elucidating of anti-inflammatory substances of Salvia plebeia lays of foundation for its quality evaluation and comprehensive development and uti-lization of resources.
