Pharmacodynamic components of Qiye Shen'an Tablets based upon spectrum-effect relationship
OBJECTIVE To explore the pharmacological substances of Qiye Shen'an Tablets(QST)in improving insomnia through spectrum-activity correlation analysis,and to provide experimental basis for the quality standards.METHODS High-performance liquid chromatography(HPLC)fingerprint of QST was established.Movement distance and duration of established zebrafish insomnia model as pharmacodynamic indicators were used.Grey correlation analysis(Grey)and partial least squares regression method(PLSR)were employed for examining the spectrum-activity relationship.UPLC-QE-Orbitrap-MS technology was utilized for detecting the related chromatographic peaks.Finally activity evaluation of screened monomers was performed.RESULTS Eleven common peaks were detected by HLPC fingerprinting of 25 batches of QST.Gray correlation analysis results indicated that all 11 common peaks were correlated with the pharmacological effect.The results of partial least squares regression analysis revealed that peak 8 contributed the most to the pharmacological effect and 10 components were identified from 11 com-mon peaks.Peak 8 was ginsenoside Rb3.Through the evaluation assay of ginsenoside Rb3,ginsenoside Rb3 possessed the effect of improving insomnia.CONCLUSION The spectrum-activity relationships of HPLC chromatogram and improving insomnia effect have been established.And the effect of QST is derived from the joint action of multiple components.A certain concentra-tion of ginsenoside Rb3 has the effect of improving insomnia.This study provides the experimental rationales for quality standard researches.
