当归补血汤提取工艺优化及其对脑缺血再灌注损伤模型大鼠的脑保护作用研究

目的:优化当归补血汤(Danggui Buxue Tang,DBT)的提取工艺,并探讨其对脑缺血再灌注损伤模型大鼠症状的改善作用。方法:以黄芪甲苷、毛蕊异黄酮、阿魏酸、毛蕊异黄酮葡萄糖苷、芒柄花素、藁本内酯6种成分含量为评价指标,采用熵权法-层次分析法(analytic hierarchy process,AHP)计算权重系数,多指标综合评分结合Box-Behnken响应面法考察溶剂浓度、料液比、提取时间对DBT提取工艺的影响;采用大鼠大脑中动脉栓塞/再灌注(middle cerebral artery occlusion/reperfusion,MCAO/R)模型评价工艺优化后的DBT对脑缺血再灌注损伤的脑保护作用。结果:最佳提取工艺条件为乙醇体积分数86%,料液比10∶1(mL∶g),提取2次,每次2。5h,9号筛过滤,减压浓缩(<50℃),冷冻干燥成冻干粉。药效学结果显示,当归补血冻干粉可改善神经功能缺损,减少脑梗死面积,减轻组织病理形态学损伤,降低大鼠脑组织中TNF-a、IL-6、IL-1β、丙二醛(malondialdehyde,MDA)的含量(P<0。05),升高过氧化氢酶(catalase assay kit,CAT)、超氧化物歧化酶(superoxide dismutase,SOD)的活性(P<0。05)。结论:优化后的DBT提取工艺科学合理、稳定可行,此工艺制得的当归补血冻干粉可改善大鼠脑缺血再灌注损伤,可为该方后续的新制剂研究提供依据。
Optimization of extraction process of Danggui BuxueTangand its protective effect on cerebral ischemia/reperfusion injury model rats
OBJECTIVE To optimize the extraction technology of Danggui Buxue Tang(DBT)and examine its improve-ment effect on the symptom of hemorrhoid model rats.METHODS The contents of astragaloside Ⅳ,calycosin,ferulic acid,calycosin-7-O-β-D-glucoside,formononetin and cis-ligustilidesix components were employed as evaluation parameters.Weight coefficient of each component was evaluated by entropy weight method-AHP.The effects of solvent concentration,solvent vol-ume and extraction time on the extraction process of DBT were investigated by multi-index comprehensive scoring combined with Box-Behnken response surface method.The middle cerebral artery occlusion/reperfusion(MCAO/R)model of rats was utilized for evaluating protective effect of the new technology of Danggui Buxue lyophilized powder on cerebral ischemia/reperfusion injury.RESULTS The optimal extraction conditions were ethanol concentration 86%,a ratio of liquid-material at 10∶1(mL∶g),extracted twice,2.5 h each time,filtered by No.10 sieve,concentrated under reduced pressure(<50 ℃)and freeze-dried into freeze-dried powder.Pharmacodynamics results showed that Danggui Buxue freeze-dried powder could improve the neurological deficit,shrink the area of cerebral infarction,alleviate the histopathological damage,reduce the contents of tumor necrosis factor-alpha(TNF-α),interleukin-6(IL-6),interleukin-1β(IL-1β)and malondialdehyde(MDA)in brain tissue of rats(P<0.05)and boost the activities of CAT and SOD(P<0.05).CONCLUSION The extraction process of DBT is scientific,reasonable,stable and feasible.The new technology of Danggui Buxue Freeze-dried Powder obtained by this process can improve cerebral ischemia/reperfusion injury in rats and provide rationales for follow-up studies of the new preparation.
