目的:建立木香药材高效液相色谱特征图谱分析方法。方法:采用Diamonsil C18色谱柱(250 mm ×4.6 mm,5μm),以甲醇( A)-0.1%甲酸水( B)为流动相梯度洗脱,流速1.0 ml/min,检测波长254 nm,柱温25℃,建立不同产地木香的高效液相色谱特征图谱;采用《中药色谱指纹图谱相似度评价系统》(2004 A版)进行相似度评价。结果:通过对15批次不同产地木香样品的分析,建立了木香药材高效液相色谱特征图谱,确定了6个共有特征峰,并指认了其中2个特征峰的结构,分别为木香烃内酯和去氢木香内酯。结论:本研究建立的木香药材特征图谱的特征性和专属性强,且方法简便、可靠,重复性好,为更好地控制木香药材的内在质量提供了科学依据。
Study on Characteristic Fingerprint of Aucklandia Radix by HPLCΔ
OBJECTIVE:To establish the HPLC characteristic fingerprint for the quality control of Aucklandia Radix. METHODS:Diamonsil C18 chromatographic column(250 mm ×4.6 mm, 5μm) was adopted, the mobile phase consists of methanol(A) and water containing 0.1%acetic acid(B) with gradient elution at the flow rate of 1.0 ml/min.The UV detection wavelength was 254 nm and the column temperature was 25 ℃.The Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine ( version 2004A) was used to calculate the similarity of the HPLC characteristic fingerprint of Aucklandia Radix from different habitats.RESULTS:HPLC characteristic fingerprint was established with six common characteristic peaks based on the analysis of 15 batches of Aucklandia Radix.Two common characteristic peaks were identified as costunolide and dehydrocostus lactone.CONCLUSIONS:The established method shows good characteristics, specificity, and repeatability, which can provide a scientific basis for the identification and quality control of Aucklandia Radix.
