Inhibitory effects and preliminary mechanism of the medicinal botanical formula MYF-20 against Malasse-zia
Objective Inhibitory effects and preliminary mechanism of the ethanol extracts of the medicinal botanical for-mula MYF-20 innovatively developed in biological laboratory against Malassezia furfur were conducted in this research.Methods The modified CLSI-M27A method was applied to determine the minimum inhibitory concentration of the formula against Malassezia furfur,and scanning electron microscopy morphology was used to learn the effect of MYF-20 on the cell integrity of Malassezia furfur,detecting the effect of the MYF-20 on the lipase activity of Malassezia furfur by method of copper soap.Moreover,infection model of Galleria mellonella was constructed to evaluate the antifungal activity of MYF-20 in vivo.Results The minimum inhibitory concentration of MYF-20 for Malassezia furfur was 7.83 mg/mL(crude dry weight of the botanical medicine drug).Furthermore,MYF-20 could significantly increase the relative conductivity of the Malassezia solution(P<0.01),scanning electron microscopy further confirmed the occurrence of spore fracture and cell rupture concavity.Meanwhile,it also significantly inhibited the activity of lipase(P<0.01).Insect experiments demonstra-ted that MYF-20 could significantly enhance the survival rate and melanization score of Galleria mellonella after infected with Malassezia and reduce the fungal burden in vivo(P<0.01).Conclusions MYF-20 has strong anti-Malassezia activity both in vitro and in vivo,and could exert its anti-Malassezia function through multiple ways,which had the potential to be developed as a new anti-Malassezia botanical drug.
