Localization investigation of molecular epidemiology of Rhodotorula mucilaginosa using rDNA sequence typing
Objective To meet the real-time molecular epidemiological investigation of Rhodotorula mucilaginosa by moni-toring units,screening efficient,rapid,and economical molecular typing methods to realize localized molecular epidemiologi-cal monitoring of clinical strains of R.mucilaginosa.Method Seventy-three clinical strains of R.mucilaginosa causing in-vasive infections were collected from 19 hospitals of China Hospital Invasive Fungal Surveillance Net (CHIF-NET) program from 2010 to 2019,as well as 21 strains isolated from environment were included.Based on different sequence characteristics on the genome,a localized molecular epidemiological surveillance method for R.mucilaginosa was screened.Result Twelve rDNA sequence types were identified based on sequences of ITS region and D1/D2 domain in rDNA.59.6% (56/94) of strains were rDNA sequence type 2,of which 92.9% (52/56) belonged to epidemic cluster strains.In total,49 different microsatellite types were identified for 73 clinical strains with 0.986 discriminatory power.The strains in the epi-demic cluster belonged to 30 microsatellite types.Conclusion The application of rDNA sequence typing can achieve localized identification of the epidemic cluster strains of R.mucilaginosa.When clinical strains of R.mucilaginosa are identified as rDNA sequence type 2,it indicates that the epidemic strains exist in the hospital environment.
