"癣净擦剂"对红色毛癣菌的体外抑菌实验研究

目的 研究土荆皮、蛇床子、大风子等9味中药醇提液对红色毛癣菌的抑菌作用,并通过扫描、透射电镜探讨其对真菌形态及超微结构的影响.方法 通过乙醇回流法提取中药用12孔板固体药基法测定9味中药对红色毛癣菌的最低抑菌浓度(MIC);通过扫描电镜、透射电镜观察单药、复方作用前后真菌细胞表面形态结构变化与细胞内超微结构变化.结果 抑菌实验示土荆皮、蛇床子、大风子、蒲公英、黄连、乌梅、黄柏、地肤子、苦参对红色毛癣菌的MIC分别为1.562 mg/mL、1.562 mg/mL、3.125 mg/mL、3.125 mg/mL、12.5 mg/mL、12.5mg/mL、25 mg/mL、50 mg/mL、>50 mg/mL;筛选出土荆皮、蛇床子、大风子等7味中药组成复方进行抑菌实验,复方MIC结果为0.545 mg/mL.扫描电镜示蛇床子、土荆皮、复方作用72 h后真菌菌丝和孢子出现不同程度的溶解、断裂和破坏.透射电镜示蛇床子、土荆皮、复方作用72 h后真菌细胞器出现不同程度的破坏和内容物流失.结论 中药复方比单药对红色毛癣菌的抑菌作用更强,单药中土荆皮、蛇床子显示出良好的抗菌效果,土荆皮、蛇床子以及复方作用后使红色毛癣菌形态和超微结构发生明显破坏和改变,破坏程度复方>土荆皮>蛇床子.
In vitro antifungal experimental study of "Xuanjing liniment" against Trichophyton rubrum
Objective To study the antifungal effect of alcohol extracts from 9 traditional Chinese medicines,including golden larch bark、Fructus cnidii、chaulmoogratree seed,etc on Trichophyton rubrum,and to explore their effects on fungal morphology and ultrastructure through scanning and transmission electron microscopy.Methods Extracting effective com-ponents of traditional Chinese medicine by ethanol reflux method;determination of the minimum inhibitory concentration (MIC) of 9 traditional Chinese medicines against Trichophyton rubrum using the 12 well plate solid drug based method;ob-serving the changes in surface morphology and intracellular ultrastructure of fungal cells before and after the action of single drug and compound drug using scanning electron microscopy and transmission electron microscopy.Results The antifungal experiment showed that the MICs of golden larch bark、Fructus cnidii、chaulmoogratree seed、dandelion、Coptis chinensis、dark plum、golden cypress、Kochia scoparia、Sophora flavescens against Trichophyton rubrum were 1.562 mg/mL,1.562 mg/mL,3.125 mg/mL,3.125 mg/mL,12.5 mg/mL,12.5 mg/mL,25 mg/mL,50 mg/mL,and>50 mg/mL,respec-tively;seven traditional Chinese medicines,including golden larch bark、Fructus cnidii、chaulmoogratree seed,etc,were se-lected to be a compound and conducted antifungal experiments,and the MIC result of the compound was 0.545 mg/mL;scanning electron microscopy showed that after 72 hours of treatment with Fructus cnidii,golden larch bark,and com-pound,the fungal hyphae and spores showed varying degrees of dissolution,breakage,and destruction;transmission elec-tron microscopy showed that after 72 hours of treatment with Fructus cnidii,golden larch bark,and compound,the fungal organelles showed varying degrees of damage and content loss.Conclusion The antifungal effect of traditional Chinese medi-cine compound on Trichophyton rubrum was stronger than any of single drug.Among the single drugs,golden larch bark and Fructus cnidii showed good antifungal effect.The morphology and ultrastructure of Trichophyton rubrum were obvi-ously damaged and changed after the action of golden larch bark、Fructus cnidii and the compound.The degrees of damage were as below:compound>golden larch bark>Fructus cnidii.
