The Discussion about the Method of Adenine Gavage Integrating High Phosphorus Dietary in Model Preparation of Chronic Kidney Disease-Mineral and Bone Disorder in Rats
Objective:To observe the adenine gavage combined with high phosphorus diet method induced chronic kidney disease-mineral and bone disorder(CKD-MBD)rat model.Methods:34 SD rats were randomly divided into control group and model group,17 rats in each group.The rats were fed with 250 mg/kg adenine suspension by gavage and 1.2%high phosphorus diet daily from the 1st to the 4th week of the experiment,and the adenine gavage was changed to once per two days from the 5th to the 8th week.The control and model groups were given equal volumes of high purity water by gavage for 8 weeks.After 8 weeks,hematoxylin-eosin(HE),periodic acid Schiff(PAS)staining and Masson staining were used to observe the renal pathology of the rats in both groups.The morphometric parameters of cortical bone morphology were measured;the levels of blood calcium(Ca),blood phosphor-us(P),blood creatinine(Scr),blood urea nitrogen(BUN)and serum whole segment parathyroid hormone(iPTH)were determined by fully automated biochemical instrument.Results:Compared with the control group,the levels of Scr,BUN,P and iPTH were sig-nificantly higher(P<0.01)and Ca levels were significantly lower(P<0.01)in the model group;the glomerular structure of the model group was disturbed,with thickened glomerular basement membrane,dilated tubules,fluid accumulation,infiltration of inflam-matory cells and brown crystalline deposits in the tubular lumen,significantly reduced renal blood vessels,interstitial fibrosis and in-flammatory cell infiltration;the model group rats had obvious calcified nodules in the aortic mesothelium with continuous linear distri-bution and local vascular rupture;the mean optical density value of α-smooth muscle actin(α-SMA)protein expression was signif-icantly decreased,and the mean optical density value of bone morphogenetic protein-2(BMP-2)and Runt-related transcription factor 2(Runx2)protein expression was increased in the model group rats,and the difference was statistically(P<0.01);increased disconnection and separation between trabecular bone networks in the proximal tibial metaphysis of rats in the model group,increased number of osteoblasts and osteoclasts on the surface of trabeculae,large amount of mesenchymal fibrosis around trabeculae,signifi-cantly lower cortical bone tissue density(P<0.01)and significantly increased cortical porosity(P<0.01)in the model group,and no difference in cortical bone volume volume fraction The differences in cortical bone volume fraction were not statistically significant(P>0.05).Conclusion:Adenine gavage combined with high phosphorus diet is a more rapid,feasible and accurate method to pre-pare CKD-MBD rat model,which is worth further promotion.
