音猬因子复合海藻酸钠水凝胶修复大鼠膝关节软骨缺损

目的:探讨音猬因子(Shh)复合海藻酸钠水凝胶对大鼠膝关节软骨缺损的修复作用。方法:培养人骨髓间充质干细胞(BMSCs),制备音猬因子复合海藻酸钠水凝胶。采用随机对照动物观察实验,选择6周龄48只健康SPF级SD雄性大鼠,全部建立膝关节软骨缺损模型,随后随机分为损伤组、观察组、对照组3组,每组16只。损伤组不进行任何处理;对照组植入不含音猬因子的海藻酸钠水凝胶25 μL与BMSCs悬液25 μL;观察组植入含音猬因子的海藻酸钠水凝胶25 μL与BMSCs悬液25 μL。12周后各组大鼠接受安乐死并观察膝关节软骨缺损和修复的效果,采用国际软骨修复协会(ICRS)评分评估,并用苏木精-伊红(HE)染色和番红O染色检测膝关节软骨组织变化,用免疫组织化学法检测各组中Ⅱ型胶原蛋白的表达,用qRT-PCR法检测各组中Sox9和糖胺多糖(GAG)的mRNA表达水平。结果:与损伤组比,对照组和观察组大鼠的ICRS评分增加,膝关节软骨组织损伤明显改善,软骨厚度明显增加,Ⅱ型胶原、Sox9 mRNA和GAG mRNA的表达水平都上调(P<0。05)。与对照组比,观察组大鼠的ICRS评分增加,膝关节软骨组织损伤明显改善,软骨厚度明显增加,Ⅱ型胶原、Sox9 mRNA和GAG mRNA的表达水平都上调(P<0。05)。结论:音猬因子复合海藻酸钠水凝胶对膝关节软骨缺损具有良好的修复作用。
Sonic Hedgehog Factor-Loaded Sodium Alginate Hydrogel on the Repair of Rat Knee Articular Cartilage Defects
Objective:To investigate the effect of sonic hedgehog factor(Shh)-loaded sodium alginate hydrogel on the repair of the rat knee articular cartilage defects.Methods:Human bone marrow mesenchymal stem cells(BMSCs)were cultured.Shh-loaded sodium alginate hydrogel was prepared.A randomized controlled animal observation experiment was conduc-ted.48 healthy SPF male SD rats aged 6 weeks were selected to establish knee cartilage defect models.They were random-ly divided into three groups:injury group,observation group,and control group,with 16 rats in each group.The injury group did not receive any treatment,the control group was implanted with 25 μL of Shh-free sodium alginate hydrogel and 25 μL of BMSCs suspension,and the observation group was implanted with 25 μL of Shh-loaded sodium alginate hydrogel and 25 μL of BMSCs suspension.After 12 weeks,the rats in each group were euthanized.and the effect of cartilage defect and repair were observed.International cartilage repair society(ICRS)scores were assigned,and histological changes in cartilage tissue were evaluated using hematoxylin and eosin(HE)staining and Safranin O staining.Immunohistoche-mistry was performed to detect the expression of type Ⅱ collagen in each group.The qRT-PCR was used to measure the mRNA expression levels of Sox9 and glycosaminoglycan(GAG)in each group.Results:Compared with the injury group,the ICRS scores of the control group and observation group rats increased,the cartilage tissue injury of knee joint was significantly improved,and the cartilage thickness was significantly increased.The expression levels of type Ⅱ collagen,Sox9 mRNA,and GAG mRNA were up-regulated in both groups(P<0.05).Compared with the control group,the ICRS scores of the observation group rats increased,the carti-lage tissue injury of knee joint was significantly improved,and the cartilage thickness was significantly increased.The expression levels of type Ⅱ collagen,Sox9 mRNA,and GAG mRNA were up-regulated in the observation group(P<0.05).Conclusion:Shh-loaded sodium alginate hydrogel exhibits excellent reparative effects on knee articular cartilage defects.
