川穹嗪在大鼠坐骨神经损伤修复中的作用及其机制研究

目的:观察川芎嗪对神经血管基膜的Ⅳ型胶原(Collagen Ⅳ)及层连蛋白(Laminin)的影响,探讨川芎嗪治疗坐骨神经损伤的作用机制。方法:Sprague Dawley(SD)大鼠随机分为假手术组(NC组)、川芎嗪组(TMP组,200 mg/kg)和生理盐水组(NS组,200 mg/kg)。以钳夹方式制作坐骨神经挤压伤模型,造模成功后采用大鼠坐骨神经功能指数(SFI)、大鼠网格爬行实验作行为学评价。进行苏木精-伊红(HE)染色,观察其病理结构形态。用免疫组化技术检测川芎嗪对坐骨神经血管基膜Ⅳ型胶原及层连蛋白表达水平的影响。结果:SD大鼠神经血管基膜HE染色及Ⅳ型胶原、层连蛋白表达部位及水平,川芎嗪组与假手术组比较差异均有统计学意义(P<0。01);川芎嗪组与生理盐水组相比,坐骨神经功能指数值有明显升高,差异有统计学意义(P<0。01)。网格实验表明,川芎嗪组与假手术组相比差异有统计学意义(P<0。01);川芎嗪组较生理盐水组踏空次数明显减少(P<0。01)。HE染色结果表明,川芎嗪组与生理盐水组相比,胶质细胞及炎性细胞增生明显。免疫组化实验结果表明,川芎嗪组和生理盐水组在大鼠坐骨神经损伤后Ⅳ型胶原、层连蛋白的表达分布于血管周围、血管内皮细胞、基膜、胶质细胞;生理盐水组的表达主要位于血管周围;川芎嗪组Ⅳ型胶原及层连蛋白表达明显增加,差异有统计学意义(P<0。05,0。01),假手术组分析结果为阴性。结论:川芎嗪能促进大鼠坐骨神经微血管基膜成分Ⅳ型胶原和层连蛋白的表达,促进血管生成,改善微循环,提升大鼠坐骨神经功能指数,治疗大鼠坐骨神经损伤。
Efficacy and Mechanism of Tetramethylpyrazine in Repair of Sciatic Nerve Injury in Rats
Objective:To explore the mechanism of tetramethylpyrazine(TMP)on the treatment of sciatic nerve injury(SNI)by regulating the basal membrane of nerve vessels.Methods:Sprague Dawley(SD)rats were randomly divided into control group,the model of sciatic nerve crush injury was made by means of hemostatic forceps.These rats were randomly divided into sham operation group(NC group),tetramethylpyrazine group(TMP group,200 mg/kg),and normal saline group(NS group,200 mg/kg).Sciatic nerve function index(SFI)and grid crawling experiments were used for behavioral evaluation.Hematoxylin-eosin(HE)staining was performed to observe its pathological structure and morphology.Immu-nohistochemistry was used to assess the expression level of Collagen Ⅳ and Laminin expression in sciatic nerve vascular basal membrane.Results:SFI results showed that there were significant differences between the two modeling groups and the NC group(P<0.01).The TMP group had a significant increase in the SFI compared with the NS group(P<0.01).The results of grid crawling experiments showed that there were significant differences between the two modeling groups and the NC group(P<0.01).The miss times in TMP group was significantly lower than that in NS group(P<0.01).The HE staining results of the TMP group showed irregular arrangement of some neuronal axons and Schwann cells,and more edema and rupture of cells compared with the NC group.The proliferation of glial cells and inflammatory cells was significantly increased in TMP group compared with the NC group.The results of immunohistochemistry showed that the expression of type Ⅳ collagen and laminin in the TMP group and NS group was distributed around the blood vessels,vas-cular endothelial cells,basal membrane and glial cells after SNI,and the expression in NS group was mainly distributed around blood vessels.The expression of type Ⅳ collagen and laminin in TMP group increased significantly compared with the NC group(P<0.05,0.01),and the analysis results in NC group were negative.Conclusion:TMP can effectively im-prove the SFI of rats,enhance the proliferation of sciatic nerve vascular endothelial cells,reduce apoptosis,promote the expression of Collagen Ⅳ and Laminin in sciatic nerve microvascular basal membrane components,thereby promoting an-giogenesis and improving lower limb function in rats after SNI.
