Study of Mechanism of the Renewing Tendon and Bone Splice Liquid in Promoting the Repair of Acute Tendon Injury
Objective:Acute tendon injury was modeled by transverse cut of the longitudinal axis of the achilles tendon in rats,and was treated by the renewing tendon and bone splice liquid to investigate the mechanism of the renewing tendon and bone splice liquid in affecting the healing of the tendon.Methods:45 SD rats were randomly divided into 5 groups:model group,sham operation group,low,medium and high dose group,with 9 rats in each group.In the sham operation group,the skin was directly sutured after incision,and the tendon injury models were established in other 4 groups and skin were sutured after tendon repair.The low,medium and high dose group were given 3.12,6.25,12.50 mL/kg of renewing tendon and bone splice liquid respectively,and model group and control group with the same amount of saline lavage intervention.The tendon healing of each group was observed 4 weeks later,and the expression of transforming growth factor β1(TGF-β1)and basic fibroblast growth factor(bFGF)in serum of each group was detected by ELISA.The expression levels of phosphatidylinositol-3 kinase(PI3K),protein kinase B(Akt)target of rapamycin(mTOR),TGF-β1 and bFGF were detected by RT-PCR.Results:Compared with model group,the expression levels of TGF-β1 and bFGF in serum of the other 4 groups were decreased(P<0.05),and the expression levels of TGF-β1,bFGF,PI3K,Akt and mTOR in tendon tissues were decreased(P<0.05).Compared with the low-dose group,the expression levels of TGF-β1 and bFGF in serum in the middle and high-dose groups were decreased(P<0.05),while the expression levels of TGF-β1,bFGF,PI3K,Akt and mTOR in tendon tissues in the middle and high-dose groups were decreased(P<0.05),and there were no significant differences in all indexes between the medium and high dose groups(P>0.05).The expression levels of TGF-β1 and bFGF in serum and TGF-β1,bFGF,PI3K,Akt and mTOR in tendon tissues were decreased in sham operation group compared with the high dose group(P<0.05).Conclusion:The renewing tendon and bone-setting liquid plays a promoting role in the process of muscle tendon healing,and can reduce tendon adhesion occurred.Its mechanism may be related to its inhibition of TGF-β1,bFGF,PI3K-Akt-mTOR pathway expression.
