Study on Mechanism of Fumigation with Zhang's Muscular Arthralgia Washing Prescription in Treating Knee Osteoarthritis
Objective:To explore the mechanism of fumigation with Zhang's muscular arthralgia washing prescription in treating knee osteoarthritis(KOA)based on Piezo mechanical sensitivity ion channel.Methods:Adult female SD rats were selected and randomly divided into blank group,model group,model+blocker group,model+TCM washing group,model+TCM washing+blocker group,respectively,with 10 animals in each group.The rats in the model group construc-ted KO A model by lower limb plaster.The model+blocker group injected GsMTx-4 on the right foot on the first day,100 μL,once a day for 7 consecutive days.The model+TCM washing group and model+TCM washing+blocker group were respectively intervened with TCM washing on the 4th day after modeling,for 20 min each time,once a day,for 21 consecutive days.Rats were killed later and synovial tissue of left hindlimb knee was taken as specimen.IL-1β,IL-6,and TNF-α in joint fluid were measured by ELISA.The protein expression of Piezo and ERK1/2 in synovial tissue of left knee was measured by Western Blot.The relative mRNA expression of Piezo and ERK1/2 was measured by RT-PCR.Results:The expression levels of Piezo two types and ERK1/2 in the knee synovial tissue of the model group rats were significantly increased compared with the blank group(P<0.05).The content of Piezo two types and ERK1/2 in the syn-ovial tissue of the left knee joint of rats in the model+blocker group was significantly reduced compared with the model group(P<0.05).The expression levels of IL-1β and IL-6 and TNF-α in the joint fluid of the knee joint cavity of the model group rats were significantly increased compared with the blank group(P<0.05).The expression levels of IL-1βand IL-6 and TNF-α in the joint fluid of rats in the knee joint cavity were significantly reduced in both model+TCM wash-ing group and model+TCM washing+blocker group compared with the model group(P<0.05).The expression levels and mRNA levels of Piezo two types and ERK1/2 in the synovial tissue of the left knee joint of the model group rats were significantly increased compared with the blank group(P<0.05).The expression levels and mRNA levels of Piezo two types and ERK1/2 in the synovial tissue of the left knee joint of rats in both model+TCM washing group and model+TCM washing+blocker group were significantly reduced compared with the model group(P<0.05).Conclusion:Zhang's muscular arthralgia washing prescription can improve the level of inflammatory factors by regulating Piezo mechanical sen-sitivity ion channel,and then alleviate the degeneration of joints.
