Chemical Constituents of Heat-clearing and Blood-cooling Decoction Analysed by LC-ESI-QTOF MS
Objective:To identify non-target compounds in heat-clearing and blood-cooling Decoction samples and to provide scientific basis for Heat-clearing and blood-cooling Decoction treating psoriasis.Methods:Ultra Performance Liquid Chromatography system equipped with a 5 cm×2.1 mm,1.7 μm BEH C18 column was used in this study.The column,constant at 40 ℃,was eluted with a linear gradient of 5%~50% B over 0~3 min,50%~ 70% B over 3~10 min,70%~100% B over 10~15 min,the composition was held at 100% B for 5 min,where A=water with 0.1% ammonium formate and B =acetonitrile.The flow rate was 0.3 mL/min.Injection volume was 1 μL for both positive ion mode(ES+) and negative ion mode(ES-).The mass spectrometric data of full scan mode was collected by using a Xevo G2 QT of MS.Results:More than 30 components were detected with relatively high abundance from the soup sample by UPLC-QTOFMS analysis.Among them,22 components were identified as citric acid (1),gallic acid (2),mudanoside B (3),catechin (6),oxypaeoniflorin (7),brevifolin carboxylic acid (8),paeonol (10),apiopaeonoside (12),albiflorin R 1 (13),paeoniflorin (14),ellagic acid (15),galloylpaeoniflorin (16),pentagalloyl glucose (17),oldenlandoside I (18),benzoyloxypaeoniflorin (19),mudanpioside C (20),benzoylpaeoniflorin(24),trihydroxy-octadecadienoic acid(25),anguivioside B1/2(26),trihydroxy-octadecenoic acid/9-Hydroxyoctadecanedioic acid (27),trihydroxy-octadecenoic acid/9-hydroxyoctadecanedioic acid (29),and 2-hydroxy-3-methylanthraquinone (30).Conclusion:Paeonol,paeoniflorin and albiflorin R1 may contribute to heat-clearing and blood-cooling Decoction.
