Effect of Sishen Pills on Expression of 5-HT Signaling Pathway in Rats with Diarrhea Irritable Bowel Syndrome
Objective:To further reveal the mechanism of visceral sensitivity in treating diarrheal irritable bowel syndrome(IBS-D)by studying the effect of Sishen Pills on the expression of 5-hydroxytryptamine(5-HT)signaling pathway in rat model of IBS-D.Methods:A total of 32 SD rats were randomly divided into normal group,model group,Sishen Pills group and dicetel group.IBS-D rat models were prepared by senna leaf gavage combined with water stress avoidance method.After 14 days of administration,bristol fecal trait scale score was performed,and visceral pain threshold was assessed according to abdominal wall withdrawal reflex.The content of 5-HT and SERT in serum was detected by ELISA,the positive expression of 5-HT in colon tissue was detected by immunohistochemistry,and the protein expression of 5-HT4R in colon tissue was detected by Western blotting.Results:Compared with normal group,the score of Bristol fecal trait scale of the model group was increased,visceral pain threshold was decreased,serum 5-HT content was increased,SERT content was decreased,positive expression of 5-HT was increased and protein expression of 5-HT4R was decreased(P<0.05 or P<0.01).Com-pared with model group,the scores of Bristol fecal trait scale decreased and visceral pain threshold increased,serum 5-HT content decreased and SERT content increased in Sishen Pills group,the positive expression of 5-HT in colon tissues was decreased and the protein expression of 5-HT4R was increased(P<0.05 or P<0.01),and the Sishen Pills group was better than dicetel group in the expression of 5-HT and 5-HT4R(P<0.05).Conclusion:The mechanism of visceral sensitivity of IBS-D treated by Sishen Pills may be related to regulating the expression of 5-HT signaling pathway.
