Objective:To explore the effect of Gualou Dahuang Decoction on lung and intestinal tissue injury in severe pneumonia rat model.Methods:A total of 60 SD rats with severe pneumonia model were randomly divided into the saline enema group and TCM enema group with 30 rats in each group,and the two groups were further di-vided into three groups respectively:the 24 h group,48 h group and 72 h group.Then 10 healthy SD rats were se-lected as the control group,and 10 severe pneumonia model rats as the model group,which were immediately exe-cuted after successful modeling,and blood,lung and colon tissues were collected.The saline enema group and the TCM enema group were given saline or TCM enema at 6 h,18 h,30 h,42 h,54 h and 66 h after successful model-ing respectively.At last,the rats were executed at 24 h,48 h and 72 h,and blood and lung and colon tissues were collected.The injury of lung and intestine was observed and scored by HE staining of pathological sections;serum surfactant protein-A(SP-A),procalcitonin and C-reactive protein were detected by ELISA;expression of SP-A and TNF-α in lung and intestine tissues were detected by Western blotting and immunofluorescence.Results:Histopathological score:The intestinal histopathological score in the saline enema group was higher than that in the TCM enema group(P<0.05);the lung histopathological score in the saline enema group had no statistical dif-ference compared with the TCM enema group(P>0.05).ELISA results showed that compared with the control group,the levels of CRP,PCT and SP-A in the model group increased(P<0.05);compared with the model group,the levels of serum CRP,PCT and SP-A in the saline enema group showed an increasing trend in each time point(P<0.05),while the levels of serum CRP and PCT in the TCM enema group did not change significantly(P>0.05),and SP-A increased from 48 h to 72 h(P<0.05).Western blotting and immunofluorescence showed the ex-pression of lung and intestine tissues SP-A increased(P<0.05)in each time point in the TCM enema group,while in the saline enema group there was no significant changes in SP-A of lung and intestine tissues(P>0.05).Western Blotting showed compared with the control group,the expression of TNF-α in lung and intestinal tissues increased in the model group.Compared with the model group,the expression of TNF-α in lung tissue at 72 h and that in intestinal tissue at 48 h,72 h decreased in the TCM enema group(P<0.05),and increased in the saline en-ema group(P<0.05)at 48 h and 72 h.Conclusion:Gualou Dahuang Decoction may reduce the damage of inflam-matory factors to lung and intestinal tissues of rats with severe pneumonia by up-regulating the expression of SP-A and down-regulating TNF-α.
