Study on the Mechanism of Sangbaipi Decoction Containing Serum in Regulating Mucus Hypersecretion in Human Bronchial Epithelial Cells
Objective:To explore the mechanism of Sangbaipi Decoction regulating mucus hypersecretion in hu-man bronchial epithelial cells.Methods:Sangbaipi Decoction containing serum and acetylcysteine solution were prepared.Human bronchial epithelial cells were treated with 15%cigarette solution and 20 ng/mL lipopolysaccha-ride solution to establish airway mucus hypersecretion model.According to drug intervention,they were divided in-to Sangbaipi Decoction group,acetylcysteine group,blank group and model group.CCK-8 method was used to de-tect the activity of human bronchial epithelial cells,electron microscope was used to observe the morphology of cells,and WB method was used to detect the protein expression of IL-1β and TNF-a predicted by data mining.Results:The experiment showed that the cell activity was decreased in the model group compared with the blank group,and the cell activity was increased in the Sangbaipi Decoction group compared with the model group(P<0.05).The protein expression of inflammatory cytokines IL-1β and TNF-α was significantly increased in the mod-el group compared with the blank group(P<0.05),indicating that the airway mucus hypersecretion model was suc-cessfully established.The protein expression of IL-1 β and TNF-α was significantly decreased in the Sangbaipi De-coction group and the acetylcysteine group compared with the model group(P<0.05).Conclusion:Sangbaipi De-coction containing serum can significantly reduce the protein expression of IL-1β and TNF-α,and its mechanism of treating airway mucus hypersecretion is related to the regulation of inflammatory cytokines IL-1β and TNF-α.
