Analgesic Effect and Immunomodulatory Mechanism of Shaoyao Gancao Decoction on Neuropathic Pain Rats through JAK2/STAT3 Signaling Pathway
Objective:To explore the analgesic effect of Shaoyao Gancao Decoction(SGD)on neuropathic pain(NP)and its mechanism of action.Methods:Spared nerve injury(SNI)was adopted to establish a NP model in rats,which were treated with SGD gavage for 14 days.The behavioral changes in rats from each group were detect-ed before and after treatment.The apoptosis of spinal dorsal horn(SDH)was detected by TUNEL staining,and im-munofluorescence staining was used to determine the co-localization of p-STAT3 and Iba1.The expression of markers associated with different phenotypes of microglia(CD68,SOCS3,Arg-1)was examined by real-time RT-PCR.The expression of IL-1 β,IL-6 and TNF-α in SDH were evaluated by ELISA.The protein expressions of JAK2、p-JAK2,STAT3,p-STAT3 in SDH were determined by Western blotting.Results:Compared with the sham group,the model group showed significant decreases on the mechanical withdrawal threshold(MWT)and thermal withdrawal latency(TWL)in the right hind paw(P<0.01),the ratio of apoptotic cells in SDH increased significantly(P<0.05),the expressions of IL-1β,IL-6,TNF-α were significantly increased(P<0.01),JAK2,p-JAK2,STAT3,p-STAT3 protein expression increased significantly(P<0.01),CD68,SOCS3 mRNA expression was increased(P<0.01),while Arg-1 mRNA expression was decreased on the postoperative 7th and 14th days(P<0.01).Compared with the model group,the SGD group showed significant increases on the MWT and TWL in the right hind paw(P<0.01),the ratio of apoptotic cells in SDH decreased with no statistical difference,the expres-sions of IL-1β,IL-6,TNF-α were significantly decreased(P<0.01),JAK2,p-JAK2,STAT3,p-STAT3 protein ex-pression decreased significantly(P<0.01),CD68,SOCS3 mRNA expression was decreased(P<0.01),while Arg-1 mRNA expression was increased on the postoperative 7th and 14th days(P<0.01).Conclusion:SGD may partici-pate in the regulation of microglial polarization in NP rats by regulating the JAK2/STAT3 signaling pathway,thus exerting its analgesic action.
