Effect of Tiaoxin Anshen Acupuncture on MicroRNAs of Myocardial Tissue of Ischemia-Reperfusion Arrhythmic Rats
Objective:To observe the effect of Tiaoxin Anshen acupuncture on the expression of miRNA(miR-NAl,miRNA133a)in myocardium of ischemia reperfusion arrhythmia(RA)model rats,and to explore the mecha-nism of ischemia reperfusion arrhythmia.Methods:RA model rats were made by ligating and loosening the anteri-or descending branch of the left coronary artery.40 Rats were divided into sham operation group,model group,acu-puncture group and amiodarone group.The acupuncture group were given Tiaoxin Anshen acupuncture pretreat-ment for 7 days at Lingtai(GV10),Shendao(GV11),bilateral Neiguan(PC6)and Baihui(GV20).After the experi-ment,ELISA method was used to detect serum CK-MB and LDH content,and HE staining was used to observe myocardial tissue,RT-PCR method was used to detect the expression levels of miRNA1 and miRNA133a in the endocardial myocardium in the central part of the ischemic area.Results:The contents of CK-MB and LDH in acupuncture group and amiodarone group were significantly lower than those in model group(P<0.01).The ex-pression of miRNA1 in acupuncture group and amiodarone group was significantly lower than that in model group(P<0.01),and amiodarone group was lower than that in acupuncture group(P<0.01).The expression of miR-NA133a in acupuncture group and amiodarone group was significantly increased compared with model group(P<0.05),but there was no difference between the two groups(P>0.05).Conclusion:Tiaoxin Anshen acupuncture pretreatment and amiodarone injection could improve myocardial damage and reduce the occurrence of ischemia-reperfusion arrhythmias,and the mechanism may be related to the downregulation of miRNA1 and upregulation of miRNA133a in myocardial tissue of ischemia-reperfusion rats.
