Exploring the Mechanism of Piantong Decoction No.1 on Rats with Pain Sensitive Migraine Based on BDNF/p-Akt Signaling Pathway
Objective:To study on the mechanisms of Piantong Decoction No.l(PTDN1)on rats with migraine induced by Nitroglycerin based on BDNF/p-Akt signaling pathway.Methods:A total of 40 SD male rats were di-vided into normal control group,sham-operated group,model control group,Zolmitriptan group and PTDN1 group by random number table method,each group had 8 rats.Rats in normal group were fed normally,rats in sham-op-erated group were injected with physiological saline,while rats in model group were injected with nitroglycerin.Each group was treated for 1 week,and the changes of behavioristics and pain threshold were detected.Blood was taken 1 h after the last dose.Enzyme-linked immunosorbent assay(ELSIA)was used to detect the protein expres-sion levels of Interleukin-6(IL-6),brain-derived neurotrophic factor(BDNF)and Nitric oxide(NO)in peripheral blood.RT-qPCR was used to detect the transcriptional level of Calclitonin gene-related peptide(CGRP),BDNF,Protein kinase B(Akt),Bcl-2-associated death promoter(BAD)and NO mRNA in Trigeminal ganglion(TG).Al-so,Western blotting was used to detect the expressions of BDNF,Akt and p-Akt protein.Results:Compared with model group,the pain thresholds of the rats in PTDN1 group were up-regulated,difference between the two groups has statistical significance(P<0.05);expression levels of BDNF,NO protein in plasma were down-regulated(P<0.05),while the expression level of IL-6 protein didn't show obvious down-regulation(P>0.05);transcriptional levels of CGRP,Akt,BDNF mRNA of rats'TG in PTDN1 group was down-regulated(P<0.05),while the tran-scriptional levels of BAD,NO mRNA didn't show obvious down-regulation(P>0.05);expression levels of Akt,p-Akt and BDNF protein in TG were clearly down-regulated(P<0.05 or P<0.001).Conclusion:The mechanism of PTDN1 in inhibiting hyperalgesia migraine is related to regulating the BDNF/p-Akt signaling pathway,down-regulating the expression levels of migraine-related factors such as CGRP,NO,and IL-6,thereby reducing the acti-vation of the pathway caused by synaptic transmitters that finally alleviate migraine attacks.
