Effects of Acupuncture with Twirling Reinforcing-Reducing Manipulation on Neurotransmitters and Inflammatory Factors in Spontaneously Hypertensive Rats
Objective:To investigate the mechanism of blood pressure regulation by observing the effects of acu-puncture with twirling reinforcing-reducing manipulation on the contents of neurotransmitters γ-aminobutyric acid(GABA)and glutamate(Glu)in the hypothalamic paraventricular nucleus(PVN),as well as the expression of in-flammatory factors interleukin-6(IL-6)and tumour necrosis factor-α(TNF-α)levels in serum of spontaneously hypertensive rats(SHR).Methods:A total of 27 male SHR were randomly assigned to twirling-reinforcing manip-ulation group,twirling-reducing manipulation group and model group,and another 9 Wistar-Kyoto(WKY)rats were used as blank group.The blank group and model group were only immobilized by grasping without acupunc-ture intervention,while acupuncture intervention was performed bilaterally on the"Taichong acupoint"in both the twirling-reinforcing manipulation group and twirling-reducing manipulation group.The groups were treated once a day in the morning for a period of 14 days,with 1 day of rest between the 2 weeks.The systolic blood pressure of the rat tail artery was measured and recorded periodically on days 0,3,7,and 14 of acupuncture.Samples were collected after the 14th day of the experiment,and the contents of GABA and Glu in the PVN tissues of rats were determined by ELISA,as well as the levels of inflammatory factors IL-6 and TNF-α in serum.Finally,histopatho-logic changes in the PVN tissues were observed by HE staining.Results:Compared with the model group,the twirling-reinforcing manipulation group and twirling-reducing manipulation group could significantly reduce the blood pressure of SHR rats(P＜0.01),and the antihypertensive effect of the twirling-reducing manipulation group was more significant(P＜0.05).After acupuncture,GABA contents were significantly increased and Glu contents were significantly decreased in the PVN tissues of the twirling-reinforcing manipulation group and twirling-reduc-ing manipulation group compared with the model group(all P＜0.01).The serum levels of IL-6 and TNF-α were significantly decreased in the twirling-reinforcing manipulation group and twirling-reducing manipulation group compared with the model group(P＜0.05 or P＜0.01).HE staining results showed that compared with the model group,the twirling-reinforcing manipulation group and twirling-reducing manipulation group improved the lesions with an increase in the total number of cells.Conclusion:Acupuncture with twirling reinforcing-reducing manipu-lation can obviously reduce blood pressure in SHR rats,and the antihypertensive effect of twirling-reducing manip-ulation group is more significant,and its mechanism of action may be related to the elevation of the neurotransmit-ter GABA contents,the reduction of Glu contents in the PVN,and the decrease in the levels of inflammatory fac-tors IL-6 and TNF-α.
