Study on the Mechanism of Dachaihu Decoction in Improving Inflammation in Rats with Severe Heat Stroke Through the TLR 4/NF-κB Pathway
Objective:To investigate the effect of Dachaihu Decoction in improving inflammation in rats with se-vere heat stroke and to investigate the mechanism of action.Methods:A total of 60 male rats were randomly di-vided into the blank control group,model group,standard dose traditional chinese medicine group and high dose traditional chinese medicine group.The classic heat stroke model was prepared by placing rats in a high tempera-ture climate animal incubator[temperature(40.50±0.50)℃,humidity(65.00±2.00)%].After successful prepara-tion,the model was administered by gavage q8h at 6 h for 2 consecutive days.5 rats were killed at 6 h,24 h and 48 h after successful modeling,and interleukin-1(IL-1),tumor necrosis factor-α(TNF-α),and ileum tissue were collected.The protein expression of TLR4 and NF-κB in intestinal tissues was determined by protein immunoblot-ting test.Results:Compared with the blank control group,the serum levels of IL-1 and TNF-α and the expres-sion levels of TLR4 and NF-κB proteins in intestinal tissues were increased in the model group,the low-dose group and the high-dose traditional chinese medicine group(P<0.05 or P<0.01).The serum levels of TNF-αand IL-1 and the protein expression levels of TLR4 and NF-κB in intestinal tissues were significantly decreased at 24 and 48 h after the model group(P<0.05).The protein expression levels of TNF-α,TLR4 and NF-κB in the high-dose traditional chinese medicine group decreased significantly at 24 and 48 h after modeling,while the lev-els of IL-1 decreased significantly at 48 h after modeling(P<0.05).Conclusion:Dachaihu Decoction can inhibit TLR 4/NF-κB signaling pathway,reduce intestinal inflammation and gastrointestinal damage,and play a role in protecting the rats with severe heat stroke.
