敦煌心痛方对心梗大鼠心肌酶、氧化应激及血清生化等因子的影响

目的 观察敦煌心痛方(硝石雄黄散)对心梗大鼠心肌酶、氧化应激及血清生化等的影响.方法 将Wistar大鼠随机分为空白组、模型组、阿司匹林组、硝酸甘油组和硝石雄黄散组.除空白组外,其余各组采用异丙肾上腺素复制急性心肌缺血大鼠模型,硝石雄黄散组使用硝石雄黄散贴敷至阳穴;硝酸甘油组和阿司匹林组分别使用硝酸甘油贴剂贴敷至阳穴和阿司匹林肠溶片灌胃;模型组和空白组给予等量生理盐水灌胃.各组大鼠于干预后1 h处死,采用HE染色观察大鼠心肌组织病理变化情况,比色法观察各干预方法对心肌酶谱肌酸激酶(CK)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)和氧化应激指标超氧化物歧化酶(SOD)、一氧化氮合酶(NOS)的影响,采用ELISA和硝酸还原酶法检测血清HMG-CoA还原酶及血管收缩因子ET、血管舒张因子NO的水平.结果 与空白组相比,模型组血清CK、AST、LDH、内皮素(ET)、NOS和HMG-CoA还原酶水平明显升高(P<0.05);与模型组相比,阿司匹林组、硝酸甘油组和硝石雄黄散组AST、LDH、ET、NOS和HMG-CoA还原酶血清水平明显降低(P<0.05),阿司匹林组和硝石雄黄散组血清CK活力明显下降(P<0.05);与阿司匹林组相比,硝酸甘油组血清LDH水平降低(P<0.05),硝酸甘油组和硝石雄黄散组ET、NOS水平上升(P<0.05);与硝酸甘油组相比,硝石雄黄散组血清CK、AST水平明显下降(P<0.05).结论 硝石雄黄散可能通过减少心肌酶的释放,改善血管内皮功能,下调NOS舒张血管,进而减轻心肌细胞缺血坏死,改善心肌梗死.
Effect of Dunhuang Xintong Formula on Myocardial Enzymes,Oxidative Stress,and Serum Biochemical Markers in Rats with Myocardial Infarction
Objective:To observe the effects of Dunhuang Xintong Formula(Xiaoshi Xionghuang Powder)on myocardial enzymes,oxidative stress,and serum biochemical markers in rats with myocardial infarction.Methods:Wistar rats were randomly divided into a control group,model group,aspirin group,nitroglycerin group,and Xiaoshi Xionghuang Powder group.Except for the control group,all other groups were subjected to acute myocardi-al ischemia induced by isoproterenol.The Xiaoshi Xionghuang Powder group received Xiaoshi Xionghuang Powder paste application on the Zhiyang acupoint(DU9),the nitroglycerin group received nitroglycerin patch,and the aspi-rin group received aspirin enteric-coated tablets.The model group and control group received an equal amount of saline orally.One hour after intervention,HE staining was used to observe pathological changes in myocardial tis-sue.Colorimetric method was used to evaluate the effects of interventions on myocardial enzyme profiles(CK,AST,LDH),oxidative stress markers(SOD,NOS),and serum levels of HMG-CoA reductase,vasoconstrictor(ET),and vasodilator(NO)factors.ELISA and nitrate reductase method were used to measure the differences in serum levels of HMG-CoA reductase,ET,and NO.Results:Compared with the control group,the model group showed signifi-cant increases in serum levels of CK,AST,LDH,ET,NOS,and HMG-CoA reductase(P<0.05).Compared with the model group,the aspirin group,nitroglycerin group,and Xiaoshi Xionghuang Powder group showed significant decreases in serum levels of AST,LDH,ET,NOS,and HMG-CoA reductase(P<0.05).The aspirin group and Xiaoshi Xionghuang Powder group exhibited significant decreases in serum CK activity compared with the model group(P<0.05).Compared with the aspirin group,the nitroglycerin group showed lower serum LDH levels(P<0.05)and higher ET and NOS levels(P<0.05).The Xiaoshi Xionghuang Powder group had significantly lower se-rum CK and AST levels compared with the nitroglycerin group(P<0.05).Conclusion:Xiaoshi Xionghuang Pow-der may attenuate myocardial ischemic necrosis,improve endothelial function,reduce the release of myocardial en-zymes,and downregulate NOS for vasodilation,thereby improving myocardial infarction.
