Improvement Effect of Oridonin on Acute Pulpitis Rats by Regulating HMGB1/TLR4 Signaling Pathway
Objective:To investigate the improvement effect of oridonin on acute pulpitis rats by regulating high mobility group protein B1(HMGB1)/Toll-like receptor 4(TLR4)signaling pathway.Methods:SD rats were divided into high dose oridonin group(20 mg/kg oridonin),low dose oridonin group(10 mg/kg oridonin),and high dose oridonin+rHMGB 1 group(recombinant protein HMGB1,intraperitoneal injection of 20 mg/kg oridonin and tail vein injection of 8 μg/kg rHMGB1),model group and control group,with 10 rats in each group.The facial comb-ing time of rats was recorded to evaluate the changes of pain behavior in rats.The levels of tumor necrosis factor(TNF-α),interleukin(IL)-1β,IL-8,IL-10,chemokine(CX3CL1)and heme oxygenase-1(HO-1)were detected by enzyme-linked immunosorbent assay(ELISA).The activity of superoxide dismutase(SOD)and the content of malondialdehyde(MD A)in dental pulp tissue were detected.The pathological changes of rat pulp tissue were ob-served by hematoxylin-eosin(HE)staining.The expression of HMGB1 mRNA and TLR4 mRNA was detected by reverse transcription polymerase chain reaction(RT-qPCR).The protein levels of HMGB 1,TLR4,nuclear factor-κB(NF-κB),p-NF-κB,B-cell lymphoma 2(Bcl-2)and Bcl-2 related X protein(Bax)were detected by Western blotting.Results:Compared with model group,the facial combing time,serum IL-1β,TNF-α,IL-8,CX3CL1,HO-1 levels,pulp tissue MDA content,HMGB 1 mRNA and TLR4 mRNA expression,HMGB 1,TLR4,p-NF-κB/NF-κB,Bax protein expression were decreased,serum IL-10 level,pulp tissue SOD activity,Bcl-2 protein expression were increased in oridonin low and high dose groups(P<0.05).Recombinant protein rHMGB1 could partially re-verse the improvement of oridonin on acute pulpitis in rats(P<0.05).Conclusion:Oridonin may reduce inflam-mation and oxidative stress in rats with acute pulpitis by inhibiting HMGB1/TLR4 signaling pathway,inhibit tissue apoptosis,reduce pulp tissue damage,and alleviate pulpitis.
