川芎嗪通过调控Txnip/TRX/NF-κB通路对急性肺损伤小鼠的保护作用研究

目的 探索川芎嗪对脂多糖(LPS)诱导的急性肺损伤(ALI)小鼠的保护作用,并探寻其可能的作用机制.方法 选取50只健康BABL/c小鼠,随机分为对照组、模型组、地塞米松组(2mg/kg)、川芎嗪低剂量组(20 mg/kg)和川芎嗪高剂量组(40 mg/kg).对照组和模型组给予生理盐水,地塞米松组(2mg/kg)、川芎嗪组(20、40 mg/kg)灌胃给予相应药物.给药1h后,除对照组外,其余各组小鼠气管滴注20 μg LPS.测定小鼠肺湿/干质量比,观察肺组织病理学变化,相应试剂盒检测肺组织超氧化物歧化酶(SOD)水平和丙二醛(MDA)含量,ELISA法检测白细胞介素-1β(1 L-1β)和肿瘤坏死因子-α(TNF-α)表达,Western blotting检测Txnip、TRX和p-NF-κB p65蛋白表达.结果 川芎嗪可显著缓解LPS诱导的ALI小鼠的肺水肿程度(P<0.01);显著上调SOD活力,下调MDA含量(P<0.05);有效降低小鼠肺组织炎性细胞因子表达水平(P<0.05);降低Txnip、p-NF-κB p65的表达,上调TRX的表达(P<0.05).结论 川芎嗪可能通过Txnip/TRX/NF-κB信号通路缓解LPS诱导的ALI.
Protective Effect of Tetramethylpyrazine on Acute Lung Injury Mice through Regulating Txnip/TRX/NF-κB Signaling Pathway
Objective:To study the protective effect of tetramethylpyrazine(TMP)on acute lung injury(ALI)in-duced by lipopolysaccharide(LPS)and to explore its possible mechanism.Methods:50 healthy BABL/c mice were randomly divided into control group,ALI group,dexamethasone(2 mg/kg)group,TMP low dose(20 mg/kg)and TMP high dose(40 mg/kg)groups.The mice were administered with saline in control and ALI groups,while dexamethasone and TMP by gavage in other groups accordingly.Except control group,the mice were given intratra-cheal instillation of 20 μg LPS 1 h after administration.The lung wet/dry weight ratio was measured,the lung his-topathological changes were observed,the corresponding kit was used to detect the level of SOD and MDA in lung tissue,the expression of 1L-1[3 and TNF-α by ELISA,and the expression of Txnip,TRX and p-NF-κB p65 pro-tein by Western blotting.Results:TMP significantly reduced the degree of pulmonary edema induced by LPS in ALI mice(P<0.01),increased the level of SOD and decreased the content of MDA(P<0.05),significantly down-regulate the levels of inflammatory cytokines in lung tissues of ALI mice(P<0.05).It decreased the protein ex-pression of Txnip and p-NF-κB p65,while increased the TRX protein expression(P<0.05).Conclusion:TMP might ameliorate the LPS-induced ALI in mice through Txnip/TRX/NF-κB signaling pathway.
