Protective Effect of Tetramethylpyrazine on Acute Lung Injury Mice through Regulating Txnip/TRX/NF-κB Signaling Pathway
Objective:To study the protective effect of tetramethylpyrazine(TMP)on acute lung injury(ALI)in-duced by lipopolysaccharide(LPS)and to explore its possible mechanism.Methods:50 healthy BABL/c mice were randomly divided into control group,ALI group,dexamethasone(2 mg/kg)group,TMP low dose(20 mg/kg)and TMP high dose(40 mg/kg)groups.The mice were administered with saline in control and ALI groups,while dexamethasone and TMP by gavage in other groups accordingly.Except control group,the mice were given intratra-cheal instillation of 20 μg LPS 1 h after administration.The lung wet/dry weight ratio was measured,the lung his-topathological changes were observed,the corresponding kit was used to detect the level of SOD and MDA in lung tissue,the expression of 1L-1[3 and TNF-α by ELISA,and the expression of Txnip,TRX and p-NF-κB p65 pro-tein by Western blotting.Results:TMP significantly reduced the degree of pulmonary edema induced by LPS in ALI mice(P<0.01),increased the level of SOD and decreased the content of MDA(P<0.05),significantly down-regulate the levels of inflammatory cytokines in lung tissues of ALI mice(P<0.05).It decreased the protein ex-pression of Txnip and p-NF-κB p65,while increased the TRX protein expression(P<0.05).Conclusion:TMP might ameliorate the LPS-induced ALI in mice through Txnip/TRX/NF-κB signaling pathway.
