Effect of Qingxuan Runmu Decoction on Ocular Surface Inflammatory Factors IFN-γ,IL-17,MMP3,and MMP9 in Dry Eye Model Mice
Objective:To investigate the effect of the traditional Chinese medicine(TCM)Qingxuan Runmu Decoction on ocular surface inflammatory factors interferon-gamma(IFN-γ),interleukin-17(IL-17),matrix metal-loproteinase 3(MMP3),and matrix metalloproteinase 9(MMP9)in dry eye model mice.Methods:Healthy female C57BL/6J mice were randomly divided into four groups,each containing 20 mice:group A(blank control group),group B(model group),group C(TCM group),and group D(hyaluronic acid group).Dry eye models were created in groups B,C,and D.Group C was treated with 18 mg/kg/day of the TCM decoction via gastric gavage twice dai-ly,and group D was treated with 0.1%hyaluronic acid eye drops twice daily.After the experiment,the lacrimal glands and corneas of the mice were collected,and immunohistochemistry was used to detect the levels of IFN-γ,IL-17,MMP3,and MMP9.Results:In the lacrimal gland tissues of the mice,compared with the B group,the ex-pression levels of IFN-γ and IL-17 proteins were decreased in the group C(P<0.05),and the expression level of MMP3 protein was significantly decreased(P<0.01).Compared with the B group,the expression level of IFN-γprotein was significantly decreased in the group D(P<0.01),and the expression level of MMP9 protein was de-creased(P<0.05).In the corneal tissues of the mice,compared with the group B,the expression level of MMP3 protein was decreased in the group C(P<0.05),and the expression level of MMP9 protein was significantly de-creased(P<0.01).Compared with the group B,the expression level of MMP9 protein was significantly decreased in the group D(P<0.01).Conclusion:Qingxuan Runmu Decoction can effectively downregulate the levels of ocu-lar surface inflammatory factors IFN-γ,IL-17,MMP3,and MMP9 in dry eye model mice,thereby alleviating ocu-lar surface damage caused by dry eyes.
