大柴胡汤对脓毒症大鼠回肠组织mRNA及非靶向代谢影响的组学研究

目的 基于mRNA高通量测序(RNA-seq)和超高效液色谱串联高分辨质谱(UPLC-QTOF-HRMS)技术观察大柴胡汤改善脓毒症大鼠肠损伤的疗效并探寻其作用机制.方法 采用盲肠结扎穿刺法建立大鼠脓毒症肠损伤(SII)模型,将90只大鼠随机分为假手术组(n=10)与模型组及大柴胡汤低、中、高剂量组(n=20),术后给药2次,24 h后观察其生存率、脓毒症严重程度(mMSS)评分;HE染色评估肠道病理改变,免疫荧光(IF)及实时荧光定量聚合酶链式反应(rt-PCR)评估肠道紧密连接蛋白Occludin的表达;生化检测肌酸激酶(CK)、乳酸脱氢酶(LDH)、肌酸激酶MB同工酶(CK-MB)、谷丙转氨酶(ALT)、谷草转氨酶(AST)、总胆红素(TBil)、尿素氮(BUN)、尿素(UREA)、肌酐(Cr);酶联免疫吸附测定试验(ELISA)评估二胺氧化酶(DAO)水平.取模型组及大柴胡汤中剂量组大鼠回肠组织做RNA-seq及非靶向代谢组学检测、分析.结果 模型组24 h生存率低于假手术组,大柴胡汤各剂量组与假手术组大鼠生存率相近(P>0.05);中剂量大柴胡汤可显著降低mMSS评分,改善肠损伤,增加Occludin表达(P<0.05).RNA-seq鉴定出205个差异mRNA,代谢组学共鉴定出968个差异代谢物.差异mRNA和差异代谢物共同富集到的通路包括:脂质消化与吸收、碳水化合物消化与吸收、蛋白消化与吸收、维生素消化与吸收,且均为正调节.此外,大柴胡汤可改善SII大鼠心、肝、肾功能.结论 大柴胡汤可降低脓毒症大鼠mMSS评分、减轻肠损伤、改善心肝肾功能,其机制可能与促进营养物质消化吸收有关.
Omics Study of the Effects of Dachaihu Decoction on mRNA Expression and Non-targeted Metabolism in Ileal Tissue of Sepsis Rats
Objective:To investigate the therapeutic effect and mechanism of Dachaihu Decoction(DCHD)on intestinal injury in septic rats based on high-throughput RNA sequencing(RNA-seq)and ultra-performance liq-uid chromatography coupled with quadrupole time-of-flight high-resolution mass spectrometry(UPLC-QTOF-HRMS).Methods:Rat model of septic intestinal injury(SII)was established using the cecal ligation and punc-ture(CLP)method.90 rats were randomly divided into a sham operation group(n=10),a model group,and low,medium,and high dose groups of DCHD(n=20,each).The drugs were administered twice postoperatively,and sur-vival rates and murine severity scores of sepsis(mMSS)were observed after 24 hours.Hematoxylin and eosin(HE)staining was used to assess intestinal pathological changes.Immunofluorescence(IF)and real-time quantitative polymerase chain reaction(rt-PCR)were used to evaluate the expression of the tight junction protein Occludin in the intestine.Biochemical tests were performed for creatine kinase(CK),lactate dehydrogenase(LDH),CK-MB iso-enzyme,alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBil),blood urea nitro-gen(BUN),urea(UREA),and creatinine(Cr).Enzyme-linked immunosorbent assay(ELISA)was used to measure diamine oxidase(DAO)levels.RNA-seq and non-targeted metabolomics analysis were conducted using ileal tis-sues from the model group and the medium dose DCHD group.Results:The 24-hour survival rate in the model group was lower than that in the sham group,while there was no significant difference in survival rates between the various DCHD dose groups and the sham group.The medium dose of DCHD significantly reduced mMSS scores,improved intestinal injury,and increased Occludin expression.RNA-seq identified 205 differentially ex-pressed mRNAs,while metabolomics identified 968 differential metabolites.These differentially expressed mRNAs and metabolites were enriched in pathways related to lipid digestion and absorption,carbohydrate digestion and ab-sorption,protein digestion and absorption,and vitamin digestion and absorption,all showing positive regulation.Ad-ditionally,DCHD improved heart,liver,and kidney function in SII rats.Conclusion:DCHD can reduce mMSS scores,alleviate intestinal injury,and improve heart,liver,and kidney functions in septic rats.The mechanism may be associated with promoting the digestion and absorption of nutrients.
