Mechanism Study on Promoting Multifidus Muscle Repair in Rats with Lumbar Disc Herniation by Triple Needling at Weizhong(BL40)
Objective:To explore the mechanism of triple needling at Weizhong(BL40)to promote the repair of multifidus muscle in rats with lumbar disc herniation.Methods:Twenty-four male SD rats aged 6-8 weeks were randomly divided into the blank group,model group,ordinary acupuncture group and triple needling group,with 6 rats in each group.In addition to the blank group,the other three groups established a rat model of intervertebral disc herniation with autologous nucleus pulposus transplantation.Acupuncture intervention started 3 days after suc-cessful modeling.The common acupuncture group was treated with common acupuncture at Weizhong(BL40),and the triple needling group was treated with triple acupuncture at Weizhong(BL40).The mechanical paw withdrawal threshold of the hind limbs of the rats was detected before modeling,1 day after modeling,3 days after modeling,7 days after acupuncture,14 days after acupuncture,and 21 days after acupuncture.HE staining was used to observe the pathological morphology of multifidus muscle.Western blotting and real-time PCR were used to detect the lev-els of VEGF,MyoD and Pax7 in multifidus muscle.Results:After modeling,the mechanical pain threshold of rats was significantly reduced.The results of HE pathological staining showed that the muscle fibers in the model group were atrophied,and the nucleus moved inward.After 21 days of acupuncture treatment,the cross-sectional area of muscle fibers increased,and the mechanical pain threshold increased significantly.The triple needling group was better than the ordinary acupuncture group(P<0.05).The levels of VEGF,MyoD and Pax7 were higher than those in the model group(P<0.05).Conclusion:Triple needling at Weizhong(BL40)can improve the pain threshold of lumbar disc herniation model rats,promote the angiogenesis of multifidus muscle and improve the abil-ity of muscle fiber repair.
