Analysis of Medication Patterns for Treating PACG in Traditional Chinese Medicine Formulations Based on Data Mining of National Patents
OBJECTIVE This study explores the medication patterns of Traditional Chinese Medicine(TCM)formulations for treating primary angle-closure glaucoma(PACG)based on data mining of national patents.METHODS Patents for Chinese medicine formulations treating PACG were included from the Chinese Patent Public Announcement website established until December 31,2022.Excel software was used to establish a database for Chinese medicine formulations,and R language was employed for frequency statistics,classification of properties and flavors,association rules,and clustering analysis.RESULTS(1)General information:A total of 160 patents were included,involving 510 medicinal ingredients with a cumulative frequency of 2,055 occurrences.(2)Classification of properties and flavors:Cold-natured medicines were the most common,with 27 flavors(49.09%).Sweet-flavored medicines were the most common among the five tastes,with 71 flavors(47.97%).The liver meridian had the highest number of medicinal ingredients(38 flavors,25.00%).(3)High-frequency use of Chinese medicines:55 high-frequency Chinese medicines(used more than 10 times)were identified,with Lycium barbarum(46 times,28.75%)being the most frequently used,followed by Rehmannia glutinosa(42 times,26.25%),Chrysanthemum morifolium(41 times,25.63%),Glycyrrhiza uralensis(39 times,24.38%),and Cassia seed(38 times,23.75%).(4)Analysis of high-frequency Chinese medicine efficacy classification:The top five efficacy classifications for high-frequency Chinese medicines were heat-clearing medicines(14 flavors,25.45%),tonifying deficiency medicines(12 flavors,21.82%),exterior-releasing medicines(eight flavors,14.55%),liver-calming and wind-extinguishing medicines(five flavors,9.09%),and diuretic and dampness-dispelling medicines(four flavors,7.27%).(5)Association rules analysis:The top five combinations were Chrysanthemum morifolium-Lycium barbarum(21 times,9.21%),Poria cocos-Lycium barbarum(20 times,8.77%),Cassia seed-Chrysanthemum morifolium(20 times,8.77%),Alisma orientale-Chrysanthemum morifolium(18 times,7.89%),and Rehmannia glutinosa-Chrysanthemum morifolium(18 times,7.89%).(6)Cluster Analysis:Six effective combinations were obtained through cluster analysis.The first category's efficacy was heat-clearing and fire-purging,liver-soothing and depression-relieving;The second category's efficacy was dispelling wind and releasing the exterior,cooling blood and detoxifying;The third category's efficacy was promoting blood circulation,dispelling stasis,promoting menstruation,and relieving pain;The fourth category's efficacy was nourishing the kidneys and replenishing Yin,calming the liver and concealing Yang;The fifth category's efficacy was nourishing the liver and kidneys,resolving phlegm,and purging fire;The sixth category's efficacy was nourishing the liver and kidney,dispelling wind,and purging fire.CONCLUSIONS There is a regular pattern in the use of TCM formulations for treating PACG in national patents.The approach mainly focuses on liver,kidney,and spleen,combining cold and warm properties,considering deficiency and excess,addressing the root cause while treating the symptoms.It primarily targets six pathological factors,including wind,fire,phlegm,deficiency,stagnation,and stasis.
