Study on the Correlation of Se-rum circCOL1A2 and miR-25-3p Levels with Type 2 Diabetes Mellitus Combined with Diabetic Retinopathy
OBJECTIVE To explore the relationship between serum circ-recombinant collagen type Ⅰ α2(COL1A2)and microRNA-25-3p(miR-25-3p)levels and diabetic retinopathy(DR)in patients with type 2 diabetes mellitus(T2DM).METHODS A total of 108 T2DM patients(216 eyes)treated at Cangzhou Central Hospital from May 2020 to June 2022 were included.They were divided into DR group(56 cases,112 eyes)and non-DR(NDR)group(52 cases,104 eyes)based on the occurrence of DR.Additionally,45 healthy individuals(90 eyes)who underwent physical examinations at the same period were selected as the control group.Peripheral venous blood samples(4~6 mL)were collected from the control group during the physical examination and from the DR and NDR groups upon admission.Real-time fluorescent quantitative polymerase chain reaction(qRT-PCR)was used to detect the relative expression levels of circCOL1A2 and miR-25-3p in the three groups,and serum biochemical indicators were measured.The correlation between serum circCOL1A2 and miR-25-3p levels,their correlation with DR occurrence,and their predictive value for DR occurrence were analyzed.RESULTS(1)Levels of circCOL1A2 and miR-25-3p:There were significant differences in serum circCOL1A2 levels among the three groups(F=685.891,P=0.000).The serum circCOL1A2 levels in the NDR and DR groups were higher than those in the control group,with statistically significant differences(qNDR group=15.969,qDR group=50.526,both P=0.000).The serum circCOL1A2 levels in the DR group were higher than those in the NDR group,with a statistically significant difference(q=35.641,P=0.000).There were also significant differences in serum miR-25-3p levels among the three groups(F=583.834,P=0.000).The serum miR-25-3p levels in the NDR and DR groups were higher than those in the control group,with statistically significant differences(qNDR group=10.101,qDR group=45.157,both P=0.000).The serum miR-25-3p levels in the DR group were higher than those in the NDR group,with a statistically significant difference(q=36.169,P=0.000).(2)Biochemical indicators:The serum C-reactive protein(CRP),uric acid(UA)levels in the DR group were higher than those in the NDR group,with statistically significant differences(tUA=3.320,P=0.001;tCRP=8.705,P=0.000).There were no statistically significant differences in other serum biochemical indicators between the two groups(P>0.05).(3)Comparison of serum circCOL1A2 and miR-25-3p levels in different stages of DR:There were significant differences in serum circCOL1A2 levels among patients with stages Ⅰ to Ⅴ of DR(F=32.900,P=0.000).The serum circCOL1A2 levels increased progressively from stage Ⅰ to Ⅴ,with statistically significant differences(qⅠ-Ⅱ=4.102,P=0.042;qⅡ-Ⅲ=4.674,P=0.014;qⅢ-Ⅳ=4.359,P=0.026;qⅣ-Ⅴ=4.347,P=0.027).There were also significant differences in serum miR-25-3p levels among patients with stages Ⅰ to Ⅴ of DR(F=32.407,P=0.000).The serum miR-25-3p levels increased progressively from stage Ⅰ to Ⅴ,with statistically significant differences(qⅠ-Ⅱ=5.206,P=0.005;qⅡ-Ⅲ=4.187,P=0.036;qⅢ-Ⅳ=4.165,P=0.037;qⅣ-Ⅴ=4.064,P=0.045).(4)Correlation analysis of circCOL1A2 and miR-25-3p levels with DR:Serum circCOL1A2 and miR-25-3p levels in T2DM patients were positively correlated with the occurrence of DR(rcircCOL1A2=0.382,rmiR-25-3p=0.479,both P=0.000).(5)Correlation analysis of serum circCOL1A2 and miR-25-3p:Serum circCOL1A2 and miR-25-3p levels in T2DM patients were positively correlated(r=0.564,P=0.000).(6)Analysis of factors related to the occurrence of DR in T2DM patients:CRP,UA,circCOL1A2,and miR-25-3p levels were risk factors for the occurrence of DR in T2DM patients.CONCLUSIONS Serum circCOL1A2 and miR-25-3p levels are upregulated in T2DM patients with DR,and both are positively correlated with the occurrence of DR.Combined detection has a certain predictive value for the occurrence of DR in T2DM patients.
