基于UHPLC-QE-MS方法结合网络药理学探讨炆何首乌抗衰老的作用机制

目的 探讨炆何首乌抗衰老的作用机制。方法 通过超高效液相色谱-质谱联用(UHPLC-QE-MS)技术,鉴定生、炆何首乌活性成分;利用相关数据库筛选出炆何首乌的主要活性成分及作用靶点,并借助注释、可视化和综合发现数据库(DAVID)开展京都基因与基因组百科全书(KEGG)富集分析以及基因本体论(GO)功能分析。结果 共得到炆何首乌有效成分82个,其中抗衰老相关疾病靶点共40个;得到丝氨酸/苏氨酸激酶1(AKT1)、表皮生长因子受体(EGFR)、缺氧诱导因子1亚基α(HIF1A)3个核心靶点,主要富集在癌症的蛋白聚糖、癌症的形成途径等信号通路上。结论 推测炆何首乌主要通过细胞凋亡过程的负调控、基因表达的负调控等途径,发挥抗衰老作用。
Study on the Anti-aging Mechanism of Stewed Polygoni Multiflori Based on UHPLC-QE-MS and Network Pharmacology
Objective To explore the anti-aging mechanism of stewed Polygonum multiflori.Methods Identifying the active ingredients of raw and fermented Polygonum multiflorum using UHPLC-QE-MS technology,screening out the main active ingredients and targets of fermented Polygonum multiflorum using relevant databases,and KEGG enrichment analysis and GO functional analysis were con-ducted using DAVID database.Results A total of 82 effective ingredients were obtained from Polygonum multiflorum,and a total of 40 targets were identified for the treatment of age-related diseases.Obtaining three core targets:AKT1,EGFR,and HIF1A,which are mainly enriched in proteoglycans in cancer and signaling pathways related to cancer formation.Conclusion Stewed Polygonum multiflo-rum mainly exerts anti-aging effects through negative regulation of cell apoptosis process and negative regulation of gene expression.
