基于UHPLC-Q-TOF-MS/MS技术的中药荆芥穗化学成分表征与鉴定

应用超高效液相色谱-四极杆飞行时间质谱联用技术(UHPLC-Q-TOF-MS/MS)对荆芥穗中的化学成分进行定性分析.采用Agilent poroshell 120 SB-C18 色谱柱(3.0 mm×100 mm,2.7 μm),以 0.1%甲酸水(A)-乙腈(B)溶液为流动相进行梯度洗脱,流速 0.4 mL·min-1,柱温 45℃,电喷雾离子源,在正、负离子模式下扫描采集数据,通过对照品比对及PeakView软件分析鉴定化合物.从荆芥穗中共鉴定出 97 个化合物,其中黄酮类化合物 28 个,酚酸类化合物 23 个,脂肪酸类化合物 23 个,萜类化合物 15 个以及其他类化合物 8 个.该研究建立的UHPLC-Q-TOF-MS/MS能快速、准确、较全面地鉴定荆芥穗药材的化学成分,为荆芥穗药效物质基础研究提供参考依据.
Characterization and identification of chemical constituents from Schizonepetae Spica based on UHPLC-Q-TOF-MS/MS technique
The chemical constituents of Schizonepetae Spica were qualitatively analyzed by UHPLC-Q-TOF-MS/MS.An Agilent po-roshell 120 SB-C18 column(3.0 mm×100 mm,2.7 μm)was used for gradient elution with 0.1%formic acid water(A)-acetonitrile(B)solution as mobile phase at the flow rate of 0.4 mL·min-1 and column temperature of 45℃.The data were collected by scanning in positive and negative ion modes,and the compounds were identified by comparison of reference materials and PeakView software.Ninety-seven compounds were identified from Schizonepetae Spica,including 28 flavonoids,23 phenolic acids,23 fatty acids,15 ter-penoids,and 8 other compounds.The UHPLC-Q-TOF-MS/MS method established in this study can identify the chemical components of Schizonepetae Spica rapidly,accurately,and comprehensively,and provide a basis for the basic study of pharmacodynamic sub-stances of Schizonepetae Spica.
