该文旨在对鲜人参Panax ginseng根中的丙二酰基人参皂苷类化学成分进行研究。通过D101 大孔吸附树脂柱、ODS柱色谱及高效液相色谱制备分离技术等方法,对鲜人参根中 70%乙醇提取物的化学成分进行分离纯化,并通过高分辨质谱和核磁共振波谱相结合的方法对化合物进行结构鉴定。从鲜人参提取物中分离得到 2 个丙二酰基人参皂苷,分别鉴定为 3-O-[6-O-丙二酰基-β-D-吡喃葡萄糖基-(1→2)-β-D-吡喃葡萄糖基]-20-O-[β-D-吡喃木糖基-(1→4)-α-L-吡喃阿拉伯糖基-(1→6)-β-D-吡喃葡萄糖基]-达玛-24-烯-3β,12β,20S-三醇(1)和 3-O-[6-O-丙二酰基-β-D-吡喃葡萄糖基-(1→2)-β-D-吡喃葡萄糖基]-20-O-[β-D-吡喃木糖基-(1→2)-α-L-呋喃阿拉伯糖基-(1→6)-β-D-吡喃葡萄糖基]-达玛-24-烯-3β,12β,20S-三醇(2)。化合物1 和 2 为首次从人参根中分离得到的新化合物,分别命名为丙二酰基人参皂苷-Ra1(malonyl ginsenoside-Ra1)和丙二酰基人参皂苷-Ra2(malonyl ginsenoside-Ra2)。
Isolation and identification of malonyl ginsenoside-Ra1 and malonyl ginsenoside-Ra2 from fresh roots of Panax ginseng
The aim of this paper is to study the malonyl ginsenosides in the fresh roots of Panax ginseng.D101 macroporous adsorption resin,ODS,and preparative HPLC were employed to separate the chemical components from the 70%ethanol extract of the fresh roots of P.ginseng,and the structures of the separated compounds were identified based on the data of high-resolution mass spectrometry and nuclear magnetic resonance spectroscopy.Two malonyl ginsenosides were isolated from the fresh roots of P.ginseng and identified as 3-O-[6-O-malonyl-β-D-glucopyranosyl-(1→2)-β-D-glucopyranosyl]-20-O-[β-D-xylopyranosyl-(1→4)-α-L-arabinopyranosyl-(1→6)-β-D-glucopyranosyl]-dammar-24-ene-3β,12β,20S-triol(1)and 3-O-[6-O-malonyl-β-D-glucopyranosyl-(1→2)-β-D-glucopyranosyl]-20-O-[β-D-xylopyranosyl-(1→2)-α-L-arabinofuranosyl-(1→6)-β-D-glucopyranosyl]-dammar-24-ene-3β,12β,20S-triol(2),respectively.Compounds 1 and 2 are new compounds isolated from fresh roots of P.ginseng for the first time and named as malonyl ginsenoside-Ra1 and malonyl ginsenoside-Ra2,respectively.
