菊花CmOMT基因家族的鉴定及分析

菊花富含羟基黄酮和甲氧基黄酮,由于甲氧基黄酮具有更好的生理活性和药理作用,因此菊花中甲氧基黄酮(OMFs)的生物合成途径解析具有重要的理论和经济价值。为了更好地促进菊花 OMFs 生物合成途径的解析,该研究基于菊花全基因组数据,采用生物信息学的方法对菊花CmOMT基因家族进行了系统分析。结果表明从菊花基因组数据库中共鉴定出 123 个CmOMT基因,编码 112~541 个氨基酸,等电点为 4。92~7。59,相对分子质量为 12 651。69~61 300。23,除CmOMT50外,其余 122 个蛋白均表现为酸性且其中 79 个蛋白为亲水蛋白。CmOMT基因主要分布在细胞质且定位分布并不均匀。KEGG分析共富集到 7 个通路,包括新陈代谢、苯丙素类生物合成、类异黄酮生物合成等;基因本体(GO)富集结果显示,123 个CmOMT基因共注释到 42 个类别,分子功能主要包括O-甲基转移酶活性、S-腺苷甲硫氨酸依赖性甲基转移酶活性等,生物过程主要包括甲基化、类黄酮代谢过程等。通过启动子分析预测,菊花CmOMT家族能响应光、干旱、低温、茉莉酸甲酯、脱落酸等多种胁迫,并有参与类黄酮生物合成基因调控的MYB结合位点。通过对菊花CmOMT基因家族的分析,为菊花OMFs的生物合成途径解析和后续候选基因的挖掘及功能验证提供了参考。
Identification and analysis of CmOMT gene family in Chrysanthemum morifolium
Chrysanthemum morifolium is rich in hydroxyflavonoids and methoxyflavonoids(OMFs),and dissecting the biosynthetic pathway of OMFs in Ch.morifolium is of great theoretical and economic value because of the diverse physiological activities and phar-macological effects of OMFs.To investigate the biosynthetic pathway of OMFs in Ch.morifolium,this study systematically analyzed the CmOMT gene family based on the whole genome data of Ch.morifolium by using bioinformatics approaches.The results showed that 123 CmOMT genes were identified in the genome of Ch.morifolium,encoding 112-541 amino acid residues.The putative proteins had the isoelectric points ranging from 4.92 to 7.59,relative molecular mass ranging from 12 651.69 to 61 300.23.Except CmOMT50,the rest 122 proteins were acidic and 79 of them were hydrophilic.CmOMTs predominantly existed in the cytoplasm with uneven distri-bution.Seven pathways were enriched by the Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis,including metabolism,phenylpropanoid biosynthesis,and isoflavonoid biosynthesis.The Gene Ontology(GO)enrichment results showed that the 123 CmOMT genes were annotated to 42 GO terms of 12 molecular functions(such as O-methyltransferase activity and S-adenosylme-thanethionine-dependent methyltransferase activity)and 27 biological processes(such as methylation and flavonoid metabolism proces-ses).The prediction results of the cis-acting elements in the promoter region suggested that the CmOMT gene family was involved in the responses to multiple biotic and abiotic stress conditions including light,drought,low temperature,methyl jasmonate,and abscisic acid.In addition,the promoters of CmOMT genes had MYB binding sites involved in the regulation of genes for flavonoid biosynthesis.The analysis of the CmOMT gene family provides a reference for deciphering the biosynthetic pathway of OMFs in Ch.morifolium and lays a foundation of the subsequent mining and functional validation of candidate OMT genes in C.morifolium.
