荷叶中生物碱类化合物及其降血脂活性研究

运用硅胶柱色谱、ODS柱色谱和制备型高效液相色谱等方法从荷叶的 70%乙醇提取物中分离提取得到 4 个生物碱类化合物,包括 1 个新化合物和 3 个已知化合物。利用核磁共振波谱、高分辨质谱、圆二色谱等方法对其进行结构鉴定,其中化合物 1 为新化合物,命名为lotustine B(1)。化合物 2~4 为已知化合物,分别鉴定为nuciferin(2)、dehydronuciferine(3)、N-methylasimilobine(4)。通过构建HepG2 细胞高血脂模型,评价了分离得到的 4 个生物碱类化合物对总胆固醇(TC)及总甘油三酯(TG)水平的影响,同时也利用油红O染色的实验方法评价了化合物 1~4 的降血脂活性,结果显示化合物 4 能降低HepG2 高脂模型细胞内的脂质累积,具有潜在的降血脂活性。
Alkaloid compounds in Nelumbinis Folium and their hypolipidemic effect
Four alkaloid compounds including one new compound and three known ones were isolated and extracted from the 70%ethanol extract of Nelumbinis Folium by silica gel column chromatography,octadecylsilyl(ODS)column chromatography and prepara-tive high-performance liquid chromatography.Their structures were identified by nuclear magnetic resonance spectroscopy,high-resolu-tion mass spectrometry,circular dichroism and other methods.Compound 1 was identified as a new compound and named as lotustine B(1).Compounds 2-4 were identified as nuciferin(2),dehydronuciferine(3),N-methylasimilobine(4).The effects of four isola-ted alkaloid compounds on total cholesterol(TC)and total triglycerides(TG)levels were evaluated by constructing a human hepatoma(HepG2)cell model of hyperlipidemia.The hypolipidemic effects of compounds 1-4 were also evaluated using oil red O staining experi-ments.The results showed that compound 4 can reduce the lipid levels of HepG2 cells and has potential hypolipidemic effects.
