Alkaloid compounds in Nelumbinis Folium and their hypolipidemic effect
Four alkaloid compounds including one new compound and three known ones were isolated and extracted from the 70%ethanol extract of Nelumbinis Folium by silica gel column chromatography,octadecylsilyl(ODS)column chromatography and prepara-tive high-performance liquid chromatography.Their structures were identified by nuclear magnetic resonance spectroscopy,high-resolu-tion mass spectrometry,circular dichroism and other methods.Compound 1 was identified as a new compound and named as lotustine B(1).Compounds 2-4 were identified as nuciferin(2),dehydronuciferine(3),N-methylasimilobine(4).The effects of four isola-ted alkaloid compounds on total cholesterol(TC)and total triglycerides(TG)levels were evaluated by constructing a human hepatoma(HepG2)cell model of hyperlipidemia.The hypolipidemic effects of compounds 1-4 were also evaluated using oil red O staining experi-ments.The results showed that compound 4 can reduce the lipid levels of HepG2 cells and has potential hypolipidemic effects.
